Das Instrument "CryoWriter" und die Protokolle erforderlich zur Vorbereitung Probe Netze für NS und Cryo-EM aus nL Größe insgesamt Probenvolumen und vollständig zu vermeiden den klassischen Papier-Blot-Schritt werden vorgestellt. Mikrofluidische Prinzipien und einem mikromechanischen System werden in der CryoWriter zu diesem Zweck kombiniert.
Unsere Erfahrung zeigt, dass wenn die miniaturisierten in diesem Manuskript vorgestellten Methoden verwendet werden, Parameterraum für EM-Grid Vorbereitung größer als bei klassischen Methoden und unter enger Kontrolle des Benutzers. Wichtig ist, macht die erhöhte Reproduzierbarkeit erreicht es möglich, vor dem Bildschirm mit Probe-Puffersystem, ergänzt mit einem leicht verfügbaren Test Protein, um die optimalen Parameter zu bestimmen, bevor das eigentliche Experiment durchgeführt wird. Dies hält Verzehr die Stichprobe von Interesse auf ein absolutes minimum und ist sehr zu empfehlen. Wichtige Schritte für beide NS und Cryo-EM Raster Vorbereitung sind: (i) grundieren des Pumpsystems; für Sub-Nanoliter Volumen Dosieren, muss System-Flüssigkeit (Wasser) entgast und bubble-kostenlos. (Ii) genaue Steuerung der Glimmentladung oder Plasma Reinigung Schritt; die Oberflächeneigenschaften des Rasters EM sind entscheidend für reproduzierbare Ergebnisse. (Iii) Probe Klimaanlage; der Zeitaufwand für die Konditionierung, z. B.mit NS, hängt von der Puffertyp, Salzgehalt und Konzentration (Abbildung 3) sowie auf die Düsengeometrie der Microcapillary24. (iv) Verdampfungsraten für NS-EM-Vorbereitungen auf eine quantitative EM; die Wirkung von Kaffee-Ring kann verbieten, die Quantitative Analyse der NS-EM-Vorbereitungen und muss unterdrückt werden, indem langsam Verdunstungsraten gesteuert durch die DP-Bühne.
Verschiedene Aspekte der vorgestellten Raster Vorbereitung Methoden sind frei kombinierbar, so dass die Entwicklung der vielseitigen Protokolle für bestimmte Proben. Typische Beispiele wären die Entfernung eines Stoffes, die hochauflösende EM, z.B., Glycerin, durch eine Klimaanlage Schritt vor dem Gitter Vorbereitung für Cryo-EM verhindert; die Einführung der Mediator Moleküle, wie Liganden durch Konditionierung vor die Gittern bereit sind; oder die Untersuchung der einzelnen Zelle lysate von NS oder Cryo-EM (Abbildung 6).
Die Verwendung von Mikrofluidik und minimales Beispiel Beträge in den vorgestellten Methoden wird Papier Löschpapier Schritte vollständig entfernt. Dies ist ein großer Vorteil weil Papier Löschpapier eine harte Behandlung für Proteine ist, potenziell Kontamination der Probe mit unerwünschten Ionen und von Natur aus führt zu massiven Probenverlust. Auf der anderen Seite werden Effekte möglicherweise verursacht durch die Luft-Wasser-Grenzfläche die dünne Probe Film gebildet, wenn Cryo-EM-Proben in klassischer Weise zubereitet werden nicht vermieden, wenn die CryoWriter verwendet wird. Gitter für Cryo-EM können mit einer weniger als 0,2 s Wartezeit zwischen Beispielanwendung und Verglasung (Daten nicht gezeigt) vorbereitet werden. Jedoch wie Proteine durch Diffusion ein paar Zehntel Nanometer in wenigen Nanosekunden Reisen, gibt es immer noch genug Zeit für sie mit der Luft-Wasser-Grenzfläche von 100 nm dicken Probe Film mehrmals zu kollidieren. Jedoch die Menge an Proteinen, die das Festhalten an der Luft-Wasser-Grenzfläche durch diese kurze Zeitlücken deutlich gesenkt werden könnte und möglicherweise verhindern, dass Protein-Denaturierung oder partikelausrichtung eingeschränkt. Ein weiterer vielversprechender Ansatz, der sensible Proteine aus der Luft-Wasser-Grenzfläche schützen könnte soll die Probe Film von niedermolekularen oberflächenaktive Stoffe abdecken. Diese Verbindungen konnten schnell durch eine Klimaanlage-Stufe in der CryoWriter vor dem Gitter Vorbereitung eingeführt werden. Die hohe Oberflächen-Volumen-Verhältnis von mikrofluidischen Systemen ist eine weitere Einschränkung der CryoWriter als Probe kann möglicherweise verloren gehen durch unspezifische Adsorption an die Microcapillary Oberfläche und Quantitative Analyse von Partikelzählung zu stören. Das Problem richtet sich auf zwei Arten: Erstens die Probe nicht reisen große Entfernungen innerhalb der Microcapillary. In der Tat bleibt das Probenvolumen Nanoliter an der Kapillare Spitze in der Verarbeitung. Zweitens die Oberfläche zum Volumen-Verhältnis ist weiter reduziert, indem mikrokapillaren mit relativ großen inneren Durchmesser, z.B., 180 µm. Dritter, die Oberflächen der mikrokapillaren können leicht passiviert werden, falls erforderlich, z. B., indem Sie sie behandeln mit handelsüblichen Polylysin Ethanol Glykole (PLL-PEG).
Die hochauflösende Analyse von Proteinen durch die EM Einzelkorn-Ansatz erfordert nur 100.000 bis einige Millionen Bilder der einzelnen eiweißteilchen. Dies bedeutet, dass mikrofluidischen Techniken genug Protein-komplexe für die strukturelle Untersuchung bereitstellen können. Eine miniaturisierte Immuno-Niederschlag-Methode für die schnelle Lokalisierung der Proteinkomplexe (ca. 1 Stunde) von minimalen Zelle Mengen (ca. 40.000 Zellen) wurde früher28entwickelt. Diese Methode wird jetzt direkt mit der miniaturisierten Probe Vorbereitungsphase von der CryoWriter verknüpft werden. Das Endziel ist eine integrierte mikrofluidische Pipeline für ultraschnelle Protein isoliert und Cryo-EM Raster Vorbereitung zu entwickeln, die weniger als zwei Stunden in allen erfordert. Darüber hinaus beweist Abbildung 6, die winzige Menge und das Volumen des Materials für die Probenvorbereitung und die fast verlustfreie Klimaanlage und Raster Vorbereitung Verfahren mit Hilfe der CryoWriter benötigt machen Sie es möglich, das Protein zu studieren komplexe einzelner Zellen. Zusammen, bilden die miniaturisierte Immuno-Niederschlag-Methode und die CryoWriter die Grundlage für eine neue Proteomik-Methode namens "Einzelzelle visuelle Proteomics", wie wir vor kurzem für Hitzeschock Experimente24gezeigt. Daten-Analyse-Algorithmen ausgerichtet auf die Analyse der "visuellen Proteomics" Bilder werden derzeit getestet.

Hard- und Software für die strukturelle Analyse von Protein-komplexe durch Transmissions-Elektronenmikroskopie (TEM) hat in den letzten Jahren massiv vorangetrieben. Die vorgenommenen Verbesserungen ebnete den Weg für eine "Entschließung Revolution"1,2 und Strukturforschung, grundlegend verändert. Die Revolution begann mit dem Aufkommen der Cryo-Elektron Mikroskopie (Cryo-EM)3,4, so dass die Vorbereitung von biologischen Proben unter physiologischen Bedingungen Strahlenempfindlichkeit abnimmt und verhindert Probe verdampfen im Hochvakuum der Getriebe-Elektronenmikroskop-5. In den folgenden Jahren stieg die inkrementellen Fortschritts allmählich die erzielbare Auflösung. Unter diesen Neuerungen waren die Anwendung der Feldemission Gewehren6,7, und in jüngerer Zeit, verbessert Daten-Analyse-Algorithmen, z. B. maximum-Likelihood-Methoden-8,-9. Direkte Elektron Detektors Kameras10,11,12,13, Filmmodus Bildgebung und der zugehörigen Software Entwicklungen14,15, 16 , 17, zur Verfügung gestellt des endgültigen Durchbruch zu atomarer Auflösung für biologische Proben vorgeschriebenen Einzelkorn Analyse (für eine Rezension siehe Cheng, Grigorieff, Et al. 18). die Bedeutung der Cryo-EM wurde vor kurzem durch die Verleihung des Nobelpreises für Chemie zu drei von den Pionieren erkannt.
Zu eine biologische Probe durch TEM image, die Methode verwendet, um das EM-Raster mit Probe (nachfolgend als "Raster Vorbereitung" genannt) zu laden muss sicherstellen, dass die daraus resultierenden Probe-Schicht (i) dünn genug ist (< 100 nm), vermeiden umfangreiche Lärm durch unelastische oder multiplizieren verstreut Elektronen, (Ii) hält die Hochvakuum des Elektronenmikroskops, und (Iii) die Biomoleküle vor Strahlenschäden schützt. Zwei Methoden werden verwendet, um diese Voraussetzungen zu erfüllen: Negative Fleck(NS)19,20 Verfahren (Abb. 1A) adsorbieren, die Probe zu einem dünnen Carbon-Film, Biomoleküle in amorphen Schwermetall einbetten und dann ermöglichen Sie die Montage an Luft trocknen. Dies ist einfach und schnell, und die geladenen EM-Raster (nachfolgend als "Probe Grids" genannt) sind einfach zu speichern und kann für längere Zeit aufbewahrt werden (in der Regel Jahre). In TEM die Vorbereitungen ausstellen, hohen Kontrast aufgrund der NS und Elektron-Dosen als Cryo-Präparate zu tolerieren, aber die Auflösung beschränkt sich auf ca. 20 Å Cryo-EM-Verfahren, die (Abbildung 1B) beschäftigen löchrigen Kohlenstoff unterstützt. Eine dünne Folie aus der Beispiellösung erstreckte sich über die Löcher und das EM-Raster stürzte sich in ein Cryogen, in der Regel flüssiges Ethan zu schnell abkühlen unter-150 ° C. Das Ergebnis ist eine amorphe, verglast, 50 bis 100 nm dicken Film der Lösung in die Löcher der Unterstützung. Dünne, amorphe Film widersteht das Hochvakuum im Elektronenmikroskop und im Idealfall, biologische Strukturen in ihrem nativen Zustand bewahrt. Es kann mit einer relativ hohen Elektron Dosis aufgrund der niedrigen Temperatur, aber der Kontrast abgebildet und Signal-Rausch-Verhältnis ist dennoch gering. Daher durchschnittlich Techniken werden eingesetzt, um den Kontrast zu erhöhen und, sofern die Probe aus verschiedenen Blickwinkeln abgebildet wird, eine hochauflösende dreidimensionale (3D) Karte rekonstruiert werden kann. Negativen Fleck TEM (NS-EM) ist wichtig für das Screening und Qualitätskontrolle, bei hohem Kontrast erforderlich ist oder wenn nur begrenzte Mengen an Probe zur Verfügung stehen (Adsorption an den Kohlenstoff-Film im Allgemeinen konzentriert sich die Probe). Einzelkorn Cryo-EM ist die Gold-Standard-Methode, wenn hochauflösende 3D Rekonstruktionen von der Proteinstruktur angestrebt werden.

Um eine dünne Probe Film zu bilden, wird die überschüssige Flüssigkeit von Papier-Blot-Gesicht-on aus ein- oder beidseitig (Ii) entfernt. Schließlich ist das Netz rasch in flüssigem Ethan für Verglasung (Iii) gestürzt. C) NS-EM Raster Vorbereitung mit dem CryoWriter-Setup: A 5 nL Volumen wird aus dem Probe-bestand mit einer Microcapillary (i) abgesaugt. Für Probe Konditionierung taucht die Microcapillary Spitze in die Klimaanlage-Lösung, z.B.2 % Ammoniumacetat. Ionen und kleine Moleküle werden durch Diffusion (Ii) ausgetauscht. Beachten Sie, dass die Abmessungen der Microcapillary sicherzustellen, dass der gesamte Prozess Diffusion angetrieben ist. Die Temperatur der Plattform wird mit einem Versatz von der Taupunkt-Temperatur der Grid-Umgebung gesteuert. Das Raster wird bezogen auf die Microcapillary mit der Probe verschoben und die Microcapillary wird abgesenkt, bis es wenige Mikrometer über dem Raster ist. Anschließend sind wenige nanoliter Probe daraus verzichtet, während die Bühne spiralförmig bewegt wird; überschüssige Flüssigkeit wird erneut abgesaugt (i). Nach EM Raster Grundierung die Microcapillary wird zurückgezogen und das Gitter bleibt auf dem kontrollierten Bahnsteig Temperatur (nachfolgend als Taupunkt (DP) Bühne genannt) für eine kurze Zeit erlauben eine kontrollierte Menge der Probe zu verdampfen. Für Sprung Einfrieren ist das Gitter schnell zurückgezogen von der Bühne mit der Pinzette (Ii), umgedreht um 90° in die vertikale Position und stürzte in ein Cryogen-Bad (Iii) (nachfolgend als "Pick-und-Sprung" Mechanismus genannt). <a href="/files/ftp_upload/57310/57310fig1large.jpg" target="_blank">Bitte klicken Sie hier für eine größere Version dieser Figur.</a>
Leider haben die Gitter Vorbereitung Methoden für NS und Cryo-EM nicht wesentlich verbessert, da sie erfunden wurden. Aktuelle Nachteile sind der hohe Sample-Verbrauch (ca. 3 µL 1 mg/mL Protein) und die große Menge der Probe (> 99 %) verloren (Abb. 1A, B). Darüber hinaus ist die klassische Methode verwendet, um Netze für Cryo-EM vorbereiten eine harte Prozedur für Proteine: zuerst, es beinhaltet einen umfangreichen Gesicht-auf Papier-Blot-Schritt (Abbildung 1B, Ii), und zweitens ist das Protein der Luft-Wasser-Grenzfläche ausgesetzt für eine erhebliche Menge an Zeit21. Die hauseigene gebaute Setup, genannt "CryoWriter", verwendet miniaturisierte Beispiel Umgang mit Technologie und mikrofluidischen Prinzipien zu Aspirieren, Zustand und verzichten Probe, Vermeidung von Papier Löschpapier komplett und bieten alternative Methoden, um dünne Proben für Cryo-EM. Es deutlich reduziert den Verbrauch der Probe und verbessert Benutzersteuerelement über Probenvorbereitung als Ganzes. Darüber hinaus ermöglicht die Methode neuartige experimentelle Anwendungen; wie die Herstellung von isolierten biologischen Komponenten einzelner Zellen in ein Konzept namens "Einzelzelle visuelle Proteomics"22,23,24,25.

Aufgrund der jüngsten technologischen Fortschritt wird Kryo-Elektronenmikroskopie (Cryo-EM) zunehmend eine Standardmethode für die strukturelle Analyse von Protein-komplexe in atomarer Auflösung. Protein Isolationstechniken und probenvorbereitungsmethoden für EM bleiben jedoch einen Engpass. Eine relativ kleine Anzahl (100.000 auf ein paar Millionen) der einzelnen eiweißteilchen müssen für die hochauflösende Analyse von Proteinen durch die einzelnen Partikel EM Ansatz, Herstellung miniaturisierter Probe Umgang mit Techniken und Prinzipien mikrofluidischen abgebildet werden machbar sind.
Eine miniaturisierte, beflecken-papierfrei EM Raster Vorbereitung Methode zur Probe Vorkonditionierung, EM Raster Grundierung und Post-Processing, die verbraucht nur Nanoliter-Volumen der Probe wird vorgestellt. Die Methode verwendet eine Dosieranlage mit Sub-Nanoliter Präzision, Kontrolle der Flüssigkeitsaufnahme und EM Raster Priming, eine Plattform, die Raster-Temperatur, wodurch die Bestimmung der relativen Luftfeuchtigkeit über das EM-Raster und einen Pick-und-Sprung-Mechanismus zur Probe zu kontrollieren Verglasung. Für Cryo-EM ein EM-Gitter befindet sich auf der Bühne mit kontrollierter Temperatur und die Probe wird in einer Kapillare abgesaugt. Die Kapillare Spitze befindet sich in der Nähe der Oberfläche, das Gitter wird mit der Probe geladen und Exzess ist neu in der Microcapillary abgesaugt. Anschließend wird die Probe Film stabilisiert und leicht verdünnt durch kontrollierte Wasserverdampfung durch den Versatz der Plattform im Vergleich zu den Taupunkt Temperatur geregelt. Zu einem bestimmten Zeitpunkt wird der Pick-Sprung-Mechanismus ausgelöst, Grundierte EM Netz in flüssigem Ethan für Probe-Verglasung schnell zu übertragen. Alternativ stehen Probe-Konditionierung Nanoliter große Probenvolumina auf negative Fleck (NS) EM vorzubereiten.
Die Methoden stark Probe Verbrauch reduzieren und vermeiden Ansätze potenziell schädliche Proteine, wie z. B. das Filterpapier Beflecken in konventionellen Methoden verwendet. A Method for Obtaining Serial Ultrathin Sections of Microorganisms in Transmission Electron Microscopy

Observing cells and cell components in three dimensions at high magnification in transmission electron microscopy requires preparing serial ultrathin sections of the specimen. Although preparing serial ultrathin sections is considered to be very difficult, it is rather easy if the proper method is used. In this paper, we show a step-by-step procedure for safely obtaining serial ultrathin sections of microorganisms. The key points of this method are: 1) to use the large part of the specimen and adjust the specimen surface and knife edge so that they are parallel to each other; 2) to cut serial sections in groups and avoid difficulty in separating sections using a pair of hair strands when retrieving a group of serial sections onto the slit grids; 3) to use a 'Section-holding loop' and avoid mixing up the order of the section groups; 4) to use a 'Water-surface-raising loop' and make sure the sections are positioned on the apex of the water and that they touch the grid first, in order to place them in the desired position on the grids; 5) to use the support film on an aluminum rack and make it easier to recover the sections on the grids and to avoid wrinkling of the support film; and 6) to use a staining tube and avoid accidentally breaking the support films with tweezers. This new method enables obtaining serial ultrathin sections without difficulty. The method makes it possible to analyze cell structures of microorganisms at high resolution in 3D, which cannot be achieved by using the automatic tape-collecting ultramicrotome method and serial block-face or focused ion beam scanning electron microscopy.

This study presents reliable and easy procedures for obtaining serial ultrathin sections of a microorganism without expensive equipment in transmission electron microscopy.

Microcrystal Electron Diffraction of Small Molecules

A detailed protocol for preparing small molecule samples for microcrystal electron diffraction (MicroED) experiments is described. MicroED has been developed to solve structures of proteins and small molecules using standard electron cryo-microscopy (cryo-EM) equipment. In this way, small molecules, peptides, soluble proteins, and membrane proteins have recently been determined to high resolutions. Protocols are presented here for preparing grids of small-molecule pharmaceuticals using the drug carbamazepine as an example. Protocols for screening and collecting data are presented. Additional steps in the overall process, such as data integration, structure determination, and refinement are presented elsewhere. The time required to prepare the small-molecule grids is estimated to be less than 30 min.

Sample Preparation and Experimental Design for In Situ Multi-Beam Transmission Electron Microscopy Irradiation Experiments

There is a need to understand materials exposed to overlapping extreme environments such as high temperature, radiation, or mechanical stress. When these stressors are combined there may be synergistic effects that enable unique microstructural evolution mechanisms to activate. Understanding of these mechanisms is necessary for the input and refinement of predictive models and critical for engineering of next generation materials. The basic physics and underlying mechanisms require advanced tools to be investigated. The in situ ion irradiation transmission electron microscope (I&#179;TEM) is designed to explore these principles.
To quantitatively probe the complex dynamic interactions in materials, careful preparation of samples and consideration of experimental design is required. Particular handling or preparation of samples can easily introduce damage or features that obfuscate the measurements. There is no one correct way to prepare a sample; however, many mistakes can be made. The most common errors and things to consider are highlighted within. The I&#179;TEM has many adjustable variables and a large potential experimental space, therefore it is best to design experiments with a specific scientific question or questions in mind.
Experiments have been performed on large number of sample geometries, material classes, and with many irradiation conditions. The following are a subset of examples that demonstrate unique in situ capabilities utilizing the I3TEM. Au nanoparticles prepared by drop casting have been used to investigate the effects of single ion strikes. Au thin films have been used in studies on the effects of multibeam irradiation on microstructure evolution. Zr films have been exposed to irradiation and mechanical tension to examine creep. Ag nanopillars were subjected to simultaneous high temperature, mechanical compression, and ion irradiation to study irradiation induced creep as well. These results impact fields including: structural materials, nuclear energy, energy storage, catalysis, and microelectronics in space environments.

Sample preparation techniques are outlined with specific considerations for in situ ion irradiation TEM experiments. Ion species, energy, and fluence are discussed with methods for how to select and compute them. Finally, procedures for conducting an experiment are described and accompanied by the representative results.

Optimizing Sample Preparation for Cryogenic Electron Microscopy

Cryogenic electron microscopy (cryo-EM) has revolutionized structural biology by enabling the study of macromolecular structures in near-native conditions, suspended in vitreous ice. This technique allows for the high-resolution visualization of proteins and other biomolecules without the need for crystallization, offering significant insights into their function and mechanism. Recent advancements in single-particle analysis, coupled with improved computational data processing, have made cryo-EM an indispensable tool in modern structural biology. Despite its growing adoption, cryo-EM faces persistent challenges that can limit its effectiveness, particularly uneven particle distribution. This issue often leads to poor resolution and reduced accuracy in reconstructed protein structures. This article outlines a simple, practical approach to address this challenge, using the small heat-shock protein from Methanocaldococcus jannaschii (MjsHSP16.5) as an example. The method optimizes sample preparation to minimize preferential adsorption, ensuring more homogeneous particle distribution and higher-quality protein cryo-EM structures. This technique offers valuable guidance for researchers aiming to overcome similar challenges in structural studies.

Optimizing Sample Preparation Process for Transmission Electron Microscopy of Neuromuscular Junctions in Drosophila Larvae

The Drosophila neuromuscular junction (NMJ) has emerged as a valuable model system in the field of neuroscience. The application of confocal microscopy at the Drosophila NMJ enables researchers to acquire synaptic information, encompassing both quantitative data on synapse abundance and detailed insights into their morphology. However, the diffuse distribution and limited visual range of the TEM present challenges for the ultrastructural analysis. This study introduces an innovative and efficient sample preparation method that surpasses the conventional approach. The procedure begins by placing a metal mesh at the base of a flat-bottomed bottle or test tube, followed by positioning fixed larvae samples onto the mesh. An additional mesh is placed over the samples, ensuring that they are positioned between the two meshes. The fixed samples are thoroughly dehydrated and infiltrated before proceeding with the embedding procedure. Then embedding of the samples in epoxy resin is performed in a flat sheet manner, which allows for the preparation of muscles for positioning and sectioning. Benefiting from these steps, all the muscles of Drosophila larvae can be visualized under light microscopy, therey facilitating subsequent positioning and sectioning. Excess resin is removed after locating the 6th and 7th muscles of body segments A2 and A3. Serial ultra-thin sectioning of the 6th or 7th muscle is performed.

An Alternative Approach for Sample Preparation with Low Cell Number for TEM Analysis

Transmission electron microscopy (TEM) provides details of the cellular organization and ultrastructure. However, TEM analysis of rare cell populations, especially cells in suspension such as hematopoietic stem cells (HSCs), remains limited due to the requirement of a high cell number during sample preparation. There are a few cytospin or monolayer approaches for TEM analysis from scarce samples, but these approaches fail to get significant quantitative data from the limited number of cells. Here, an alternative and novel approach for sample preparation in TEM studies is described for rare cell populations that enables quantitative analysis.
A relatively low cell number, i.e., 10,000 HSCs, was successfully used for TEM analysis compared to the millions of cells typically used for TEM studies. In particular, Evans blue staining was performed after paraformaldehyde-glutaraldehyde (PFA-GA) fixation to visualize the tiny cell pellet, which facilitated embedding in agarose. Clusters of numerous cells were observed in ultra-thin sections. The cells had a well preserved morphology, and the ultra-structural details of the Golgi complex and several mitochondria were visible. This efficient, easy and reproducible protocol allows sample preparation from a low cell number and can be used for qualitative and quantitative TEM analysis on rare cell populations from limited biological samples.

Preparation and Observation of Thick Biological Samples by Scanning Transmission Electron Tomography

This report describes a protocol for preparing thick biological specimens for further observation using a scanning transmission electron microscope. It also describes an imaging method for studying the 3D structure of thick biological specimens by scanning transmission electron tomography. The sample preparation protocol is based on conventional methods in which the sample is fixed using chemical agents, treated with a heavy atom salt contrasting agent, dehydrated in a series of ethanol baths, and embedded in resin. The specific imaging conditions for observing thick samples by scanning transmission electron microscopy are then described. Sections of the sample are observed using a through-focus method involving the collection of several images at various focal planes. This enables the recovery of in-focus information at various heights throughout the sample. This particular collection pattern is performed at each tilt angle during tomography data collection. A single image is then generated, merging the in-focus information from all the different focal planes. A classic tilt-series dataset is then generated. The advantage of the method is that the tilt-series alignment and reconstruction can be performed using standard tools. The collection of through-focal images allows the reconstruction of a 3D volume that contains all of the structural details of the sample in focus.

Sample Preparation and Imaging of Exosomes by Transmission Electron Microscopy

Exosomes are nano-sized extracellular vesicles secreted by body fluids and are known to represent the characteristics of cells that secrete them. The contents and morphology of the secreted vesicles reflect cell behavior or physiological status, for example cell growth, migration, cleavage, and death. The exosomes&#39; role may depend highly on size, and the size of exosomes varies from 30 to 300 nm. The most widely used method for exosome imaging is negative staining, while other results are based on Cryo-Transmission Electron Microscopy, Scanning Electron Microscopy, and Atomic Force Microscopy. The typical exosome&#39;s morphology assessed through negative staining is a cup-shape, but further details are not yet clear. An exosome well-characterized through structural study is necessary particular in medical and pharmaceutical fields. Therefore, function-dependent morphology should be verified by electron microscopy techniques such as labeling a specific protein in the detailed structure of exosome. To observe detailed structure, ultrathin sectioned images and negative stained images of exosomes were compared. In this protocol, we suggest transmission electron microscopy for the imaging of exosomes including negative staining, whole mount immuno-staining, block preparation, thin section, and immuno-gold labelling.

Transmission electron microscopy (TEM) is characterized by high resolution and a respectable history elucidating retinal development and synapse maturation in humans and various species. It is a powerful tool to explore synaptic structure in ROs and is widely used in the research field of ROs. Therefore, to better explore the structure of RO synaptic contacts at the nanoscale and obtain high-quality microscopic evidence, we developed a simple and repeatable method of RO TEM sample preparation. This paper describes the protocol, reagents used, and detailed steps, including RO fixation preparation, post fixation, embedding, and visualization.
