Quando este método é realizado, existem algumas etapas essenciais que devem ser consideradas ao seguir o protocolo. Um passo crítico é a etapa de limpeza com solução ácida de piranha. Passo 2.1 não deve ser mais de 10 min. A solução de modelo para a etapa 1.4 não deve exceder 0,1 mg/mL, desde que estes valores foram previamente otimizados. Exames titulimétricos cíclica não devem exceder 15 ciclos a fim de obter a espessura ideal. Para etapa 3.1.3, 1,5 µ l é um valor otimizado. Este valor não deve ser mais elevado para este tipo específico de eletrodo. Se o sistema de cura UV tem uma potência de 400 W, a polimerização deve ser executada por um período máximo de 10-15 min. Tão logo o APS (iniciador) é adicionado à solução após TEMED, a etapa subsequente deve ser executada muito rapidamente para evitar a polimerização imediata (passo 3.2.5).
Um dos passos mais importantes é a remoção do carimbo modelo da superfície após a polimerização UV. Se esta etapa não é realizada corretamente, há um risco de que o filme polimérico na superfície do eléctrodo pode ser removido com o carimbo. Portanto, é recomendável para mergulhar o eléctrodo e o carimbo de proteína no topo de uma solução de água após a polimerização e, em seguida, retire o selo muito lentamente e com cuidado a superfície (etapa 3.1.6).
Baseado no modelo utilizado, modificações no tipo e proporção de monômeros usados (monómero funcional e cross-linker) poderão ser avaliadas em termos de geração de maior sensibilidade. Isso deve ser determinado empiricamente. Além disso, a ligação de afinidade da molécula modelo geralmente envolve várias interações diferentes simultaneamente. Portanto, isso pode levar a problemas durante as etapas de regeneração. Se o modelo vinculado não é liberado da superfície corretamente, isso pode influenciar a capacidade de reutilização do eletrodo para posterior análise. Essas afinidades multipontos também podem resultar de ligação através de interações fracas. Em tais sistemas, inespecificas podem ter lugar, que pode influenciar negativamente o a seletividade do sistema16. Estas são comuns e gerais, limitações do método.
Além dessas limitações específicas, existem muitas vantagens significativas do método discutido sobre os métodos existentes. Enquanto RIAs, Elisa e medições fluorométrica são muito sensíveis, eles exigem o uso de material rotulado (modelo ou detector), enquanto o biosensor é completamente rótulo livre. Esses métodos também são mais caros e demorados. Uma abordagem de biosensor permite a síntese rápida, paralelo de MIPs em composições diferentes, o mesmo tempo16. Uma vez que apenas alguns microlitros de solução de monômero é necessária para a preparação, o método é conveniente quando usando caros ou não limitado de monômeros. Além disso, o único eletrodos MIP podem ser usados para aproximadamente 80 análises sem uma diminuição significativa no desempenho, que é significativamente maior do que outros de métodos existentes30. Os métodos existentes também sofrem, em graus variáveis, de baixa sensibilidade e seletividade, enquanto o método descrito permite para detecção e quantificação de moléculas na faixa de pM com alta seletividade.
Devido a relação custo-eficácia, a facilidade de operação do instrumento e a detecção sensível em tempo real em um curto período de tempo em comparação com os métodos existentes, os biosensores são muito promissor ponto-de sistemas de deteção de cuidados sob condições de campo; por exemplo, para monitoramento ambiental e para aplicações em países em desenvolvimento. Em muitas aplicações no diagnóstico da doença, detecção em tempo real, sensível, seletiva e rápida de um biomarcador em uma mistura complexa como o soro é necessário15,25. Aqui, biosensores são superiores aos métodos existentes, em particular, devido à sua sensibilidade e robustez. Especificamente para a detecção de agentes infecciosos, bacteriófagos são recentemente considerados como elemento de biorecognition alternativos para biossensores devido à sua especificidade anfitrião de bactérias33,34,35. Substituição de anticorpos com bacteriófagos é muito promissor, a fim de reduzir o custo e aumentar a estabilidade ainda mais36. Esse sistema também permitirá a detecção e quantificação do fago específico no ambiente e de amostras clínicas. Devido a prevalência de bacteriófagos e sua capacidade de transduce bactérias com genes de resistência aos antibióticos37,38, tal método pode ser valioso para estudar a propagação de bactérias resistentes.

A quantificação das biomoléculas é usada em muitos campos diferentes de pesquisa dentro da ciência, empregando métodos como o radioimunoensaio (RIA) ou ELISA1. Alguns desses métodos requerem um rotulado reagente como um radioisótopo ou uma enzima chamada anticorpo/antigénio, o que os torna trabalhosa e demorada com procedimentos complexos2. Além disso, a robustez, seletividade e sensibilidade desses métodos não são suficientes para todas as análises; em particular não são suficientes quando quantidades attogram precisam ser analisado, ao invés de pictograma quantidades3. Para este efeito, biosensores ganharam considerável interesse4,5, em especial em combinação com impressão molecular para uma maior robustez.
Impressão molecular baseia-se na criação de cavidades, polimerização de monômeros funcionais em torno do modelo6, criação de sites de reconhecimento artificial que lembram perfeitamente o modelo7. Esta técnica tem sido usada para diversas aplicações, incluindo sistemas de entrega de droga e separação analítica, mas também como elementos biorecognition em biosensores8,9,10. No entanto, existem ainda algumas dificuldades na concepção de polímeros molecularmente impressos (MIPs) para modelos macromoleculares como proteínas e células de11,12. Devido a isto, muitos pesquisadores concentraram-se na impressão a proteína modelo diretamente sobre um substrato, criando assim uma superfície que será reconhecida pela proteína alvo12. Esta técnica de revestimento de superfície usada para empregar cavidades de reconhecimento por grandes moléculas e montagens, incluindo proteínas é chamada microcontact impressão13,14. O procedimento geral do método depende da polimerização entre duas superfícies - um selo de modelo e um suporte de polímero - sobre a qual o modelo é adsorvido em uma superfície de15,16e trouxe em contato com o superfície tratada com monômero. Por esta maneira, uma película fina de polímero é formada com o apoio através de UV-polimerização. Finalmente, o modelo é removido, deixando para trás a cavidades específicas do modelo na superfície do eletrodo impresso. Este método tem algumas vantagens, incluindo uma perda reduzida atividade da molécula imprimida, quantidades muito pequenas, bem como exigindo de moléculas modelo para a impressão processam16,17. Assim, estas superfícies cost-effective, estáveis, sensíveis e seletivas podem ser criados nas superfícies de sensor, visando a qualquer modelo de escolha do usuário.
O biosensor pode ser usado para a detecção de proteínas simples e macromoléculas muito maiores, incluindo vírus. Um grupo específico de vírus ganhando interesse recente é o bacteriófago, que é um vírus que infecta bactérias. Detecção rápida e sensível de bacteriófagos é importante durante biotecnológicas e biofarmacêutica processos a fim de determinar as infecções de culturas bacterianas com bacteriófagos18. O ensaio biológico mais comumente usado para a deteção do bacteriófago é a dupla camada agar método19, que é trabalhoso e demorado. Várias tentativas foram feitas para desenvolver novas ferramentas de diagnóstico para vírus (incluindo bacteriófagos) como força atômica (AFM) a microscopia20, interferometria21, eletroquímica22e sensor sistema23, 24. um monte de trabalho tem sido focado em biosensores devido a suas vantagens como sendo capaz de medição em tempo real,15,25, altamente sensível e fácil de operar. Um tipo específico de biosensor baseia-se em mudanças na capacitância. Estes biosensores capacitivos são os sensores eletroquímicos que medem mudanças nas propriedades dielétricas quando um analito interage com um elemento de biorecognition na superfície do sensor, causando uma diminuição da capacitância2,4 . Biosensores capacitivos têm sido utilizados para a detecção de vários analitos como antígenos, anticorpos, proteínas e heavy metal íons6,26,,27,28. Estes tipos de biosensores têm muitas vantagens como inerente rapidez, alta sensibilidade, simplicidade, baixo custo, fácil manipulação e medição em tempo real sem rotulagem29.
O método descrito neste documento destina-se a permitir a detecção e quantificação de baixo-abundantes biomoléculas em amostras altamente complexas, sem a necessidade de usar qualquer rotulagem. Em particular, a técnica é mais útil na faixa de atto-picograma de biomoléculas, onde outros instrumentos comercialmente existentes não conseguem dosar com precisão seu alvo.

ultrasensitive detection of biomarkers by using a molecular imprinting based capacitive biosensor

A capacidade de detectar e quantificar biomoléculas em soluções complexas sempre foi muito procurada dentro da ciência natural; sendo usado para a detecção de biomarcadores, contaminantes e outras moléculas de interesse. Uma técnica comumente utilizada para este fim é a enzima-lig da imunoabsorção ensaio (ELISA), onde muitas vezes um anticorpo é direcionado a uma molécula-alvo específico, e um segundo anticorpo marcado é utilizado para a detecção do anticorpo primário, permitindo a quantificação absoluta da biomolécula sob estudo. No entanto, o uso de anticorpos como elementos de reconhecimento limita a robustez do método; como faz a necessidade do uso de moléculas rotuladas. Para superar essas limitações, impressão molecular foi implementado, criação de sites de reconhecimento artificial complementar à molécula de modelo e obsoleting a necessidade do uso de anticorpos para ligação inicial. Além disso, para maior sensibilidade, o anticorpo secundário marcado pode ser substituído por biosensores depender a capacitância para a quantificação da molécula alvo. Neste protocolo, descrevemos um método para rapidamente e sem rótulo detectar e dosar baixo-abundantes biomoléculas (proteínas e vírus) em amostras complexas, com uma sensibilidade que é significativamente melhor do que os sistemas de deteção comumente usados, tais como o ELISA. Isto é tudo mediado por impressão molecular em combinação com um biossensor de capacitância.

Seguindo o protocolo, de acordo com o esquema na Figura 1, um eletrodo de ouro nua será gravado com um modelo, que representa a estrutura de um biomacromolecule. Este eletrodo pode ser aplicado em um biossensor capacitivo (Figura 2), permitindo que o aplicativo estável de um modelo para o eletrodo e a medição de mudanças na capacitância mediante vinculação do modelo.
Uma representação esquemática do biosensor capacitivo é mostrada na Figura 2. A bomba de centris, que é responsável pela injeção contínua de buffer de execução (fosfato 10 mM, pH 7,4) e o buffer de regeneração (25mm glicina-HCl, pH 2.5) durante a regeneração dentro da célula de fluxo, pode ser vista claramente na figura. A célula de fluxo consiste em trabalhar, referência e eletrodos de contador. A válvula de injeção é composta de soluções padrão de proteína/bacteriófago, que estão de passagem o desgaseificador primeiro e então injetado sequencialmente no sistema. Tão logo as soluções atingem o eletrodo de trabalho inserido na célula de fluxo, o resultado é monitorado em tempo real. Os valores de capacitância podem ser registrados, seguindo o sensorgrams na tela do computador.
Figura 3 e Figura 4 mostram as diferenças entre a superfície dos eléctrodos de ouro desencapados e imprimidas. Esta etapa de caracterização é importante para garantir que existem cavidades poliméricas, vistas como rugosidade na superfície do eléctrodo após a programação. Além de SEM, existem também outros métodos de caracterização incluindo AFM, medições de ângulo de contato, etc., que podem ser usados para caracterizar a superfície após a programação de elipsometria. Desta forma, pode ser assegurado que o processo de impressão é bem sucedido e que as cavidades modelo são formadas na superfície. Dentro dessas cavidades, o modelo pode vincular com afinidade e alta especificidade.
Depois de injectar das soluções-padrão do sistema capacitivo, uma média das cinco últimas leituras foi calculada automaticamente pelo software, e os gráficos de calibração foram obtidos plotando a mudança na capacitância vs. a concentração do analito. A diminuição da capacidade registrada surgiu a ligação do modelo. As mais moléculas que se ligam ao eléctrodo de ouro de superfície, maior a redução da capacidade total, de acordo com o princípio geral de medição capacitiva. Figura 5 e Figura 6 mostram que com o aumento da concentração do analito, a ΔC aumenta conforme o esperado. A gama dinâmica (entre os quais é o intervalo de concentração onde o sistema é útil para a detecção de um alvo específico) e o limite de detecção (LOD) podem ser avaliadas através da análise desses gráficos. De acordo com a Figura 6, o biosensor capacitivo do bacteriófago imprimido pode detectar bacteriófagos na faixa de concentração de 101 - 105 pfu/mL, com um valor LOD de pfu/mL 10 neste estudo. Tanto a Figura 5 e Figura 6 também destacar a necessidade de medir a curva de calibração no mesmo intervalo necessário para a concentração de modelo deve, desde que a linearidade da regressão pode variar mais as concentrações ( Figura 5), ou ter inclinações diferentes (Figura 6). Também note que devido as baixas concentrações utilizadas, o sistema é bastante sensível às flutuações (turbidez da amostra, projecto de ar, etc.), e portanto, é recomendável executar pelo menos triplica para reduzir o potencial de outliers, incluindo . Pela mesma razão, o desvio-padrão pode ser bastante significativo para amostras altamente diluídas, como visto na Figura 6.

Combination of Adhesive-tape-based Sampling and Fluorescence in situ Hybridization for Rapid Detection of Salmonella on Fresh Produce

This protocol describes a simple approach for adhesive-tape-based sampling of tomato and other fresh produce surfaces, followed by on-tape fluorescence in situ hybridization (FISH) for rapid culture-independent detection of Salmonella spp. Cell-charged tapes can also be placed face-down on selective agar for solid-phase enrichment prior to detection. Alternatively, low-volume liquid enrichments (liquid surface miniculture) can be performed on the surface of the tape in non-selective broth, followed by FISH and analysis via flow cytometry. To begin, sterile adhesive tape is brought into contact with fresh produce, gentle pressure is applied, and the tape is removed, physically extracting microbes present on these surfaces. Tapes are mounted sticky-side up onto glass microscope slides and the sampled cells are fixed with 10% formalin (30 min) and dehydrated using a graded ethanol series (50, 80, and 95%; 3 min each concentration). Next, cell-charged tapes are spotted with buffer containing a Salmonella-targeted DNA probe cocktail and hybridized for 15 - 30 min at 55&deg;C, followed by a brief rinse in a washing buffer to remove unbound probe. Adherent, FISH-labeled cells are then counterstained with the DNA dye 4',6-diamidino-2-phenylindole (DAPI) and results are viewed using fluorescence microscopy. For solid-phase enrichment, cell-charged tapes are placed face-down on a suitable selective agar surface and incubated to allow in situ growth of Salmonella microcolonies, followed by FISH and microscopy as described above. For liquid surface miniculture, cell-charged tapes are placed sticky side up and a silicone perfusion chamber is applied so that the tape and microscope slide form the bottom of a water-tight chamber into which a small volume (&le; 500 &mu;L) of Trypticase Soy Broth (TSB) is introduced. The inlet ports are sealed and the chambers are incubated at 35 - 37&deg;C, allowing growth-based amplification of tape-extracted microbes. Following incubation, inlet ports are unsealed, cells are detached and mixed with vigorous back and forth pipetting, harvested via centrifugation and fixed in 10% neutral buffered formalin. Finally, samples are hybridized and examined via flow cytometry to reveal the presence of Salmonella spp. As described here, our "tape-FISH" approach can provide simple and rapid sampling and detection of Salmonella on tomato surfaces. We have also used this approach for sampling other types of fresh produce, including spinach and jalape&ntilde;o peppers.

'Bioluminescent' Reporter Phage for the Detection of Category A Bacterial Pathogens

Yersinia pestis and Bacillus anthracis are Category A bacterial pathogens that are the causative agents of the plague and anthrax, respectively 1. Although the natural occurrence of both diseases&#39; is now relatively rare, the possibility of terrorist groups using these pathogens as a bioweapon is real. Because of the disease&#39;s inherent communicability, rapid clinical course, and high mortality rate, it is critical that an outbreak be detected quickly. Therefore methodologies that provide rapid detection and diagnosis are essential to ensure immediate implementation of public health measures and activation of crisis management.
Recombinant reporter phage may provide a rapid and specific approach for the detection of Y. pestis and B. anthracis. The Centers for Disease Control and Prevention currently use the classical phage lysis assays for the confirmed identification of these bacterial pathogens 2-4. These assays take advantage of naturally occurring phage which are specific and lytic for their bacterial hosts. After overnight growth of the cultivated bacterium in the presence of the specific phage, the formation of plaques (bacterial lysis) provides a positive identification of the bacterial target. Although these assays are robust, they suffer from three shortcomings: 1) they are laboratory based; 2) they require bacterial isolation and cultivation from the suspected sample, and 3) they take 24-36 h to complete. To address these issues, recombinant &#34;light-tagged&#34; reporter phage were genetically engineered by integrating the Vibrio harveyi luxAB genes into the genome of Y. pestis and B. anthracis specific phage 5-8. The resulting luxAB reporter phage were able to detect their specific target by rapidly (within minutes) and sensitively conferring a bioluminescent phenotype to recipient cells. Importantly, detection was obtained either with cultivated recipient cells or with mock-infected clinical specimens 7.
For demonstration purposes, here we describe the method for the phage-mediated detection of a known Y. pestis isolate using a luxAB reporter phage constructed from the CDC plague diagnostic phage &#934;A1122 6,7 (Figure 1). A similar method, with minor modifications (e.g. change in growth temperature and media), may be used for the detection of B. anthracis isolates using the B. anthracis reporter phage W&#946;::luxAB 8. The method describes the phage-mediated transduction of a biolumescent phenotype to cultivated Y. pestis cells which are subsequently measured using a microplate luminometer. The major advantages of this method over the traditional phage lysis assays is the ease of use, the rapid results, and the ability to test multiple samples simultaneously in a 96-well microtiter plate format.

A simple method for the identification of priority bacterial pathogens is to use genetically engineered reporter phage. These reporter phage, which are specific to their particular host species, are capable of rapidly transducing a bioluminescent signal response to host cells. Herein, we describe the use of reporter phage for the detection of Yersinia pestis.

Detection of Toxin Translocation into the Host Cytosol by Surface Plasmon Resonance

AB toxins consist of an enzymatic A subunit and a cell-binding B subunit1. These toxins are secreted into the extracellular milieu, but they act upon targets within the eukaryotic cytosol. Some AB toxins travel by vesicle carriers from the cell surface to the endoplasmic reticulum (ER) before entering the cytosol2-4. In the ER, the catalytic A chain dissociates from the rest of the toxin and moves through a protein-conducting channel to reach its cytosolic target5. The translocated, cytosolic A chain is difficult to detect because toxin trafficking to the ER is an extremely inefficient process: most internalized toxin is routed to the lysosomes for degradation, so only a small fraction of surface-bound toxin reaches the Golgi apparatus and ER6-12.

To monitor toxin translocation from the ER to the cytosol in cultured cells, we combined a subcellular fractionation protocol with the highly sensitive detection method of surface plasmon resonance (SPR)13-15. The plasma membrane of toxin-treated cells is selectively permeabilized with digitonin, allowing collection of a cytosolic fraction which is subsequently perfused over an SPR sensor coated with an anti-toxin A chain antibody. The antibody-coated sensor can capture and detect pg/mL quantities of cytosolic toxin. With this protocol, it is possible to follow the kinetics of toxin entry into the cytosol and to characterize inhibitory effects on the translocation event. The concentration of cytosolic toxin can also be calculated from a standard curve generated with known quantities of A chain standards that have been perfused over the sensor. Our method represents a rapid, sensitive, and quantitative detection system that does not require radiolabeling or other modifications to the target toxin.

In this report, we describe how surface plasmon resonance is used to detect toxin entry into the host cytosol. This highly sensitive method can provide quantitative data on the amount of cytosolic toxin, and it can be applied to a range of toxins.

Detection of Human Leukocyte Antigen Biomarkers in Breast Cancer Utilizing Label-free Biosensor Technology

According to the American Cancer Society, more than 200,000 women will be diagnosed with invasive breast cancer each year and approximately 40,000 will die from the disease. The human leukocyte antigen (HLA) class I samples peptides derived from proteasomal degradation of cellular proteins and presents these fragments on the cell surface for interrogation by circulating cytotoxic T lymphocytes (CTL). Generation of T-cell receptor mimic (TCRm) monoclonal antibodies (mAbs) which recognize breast cancer specific peptide/HLA-A*02:01 complexes such as those derived from macrophage migration inhibitory factor (MIF19-27) and NY-ESO-1157-165 enable detection and destruction of breast cancer cells in the absence of an effective anti-tumor CTL response. Intact class I HLA/peptide complexes are shed by breast cancer cells and represent potentially relevant cancer biomarkers. In this work, a breakthrough biomarker screening system for cancer diagnostics incorporating T-cell receptor mimic monoclonal antibodies combined with a novel, label-free biosensor utilizing guided-mode resonance (GMR) sensor technology is presented. Detection of shed MIF/HLA-A*02:01 complexes in MDA-MB-231 cell supernatants, spiked human serum, and patient plasma is demonstrated. The impact of this work could revolutionize personalized medicine through development of companion disease diagnostics for targeted immunotherapies.

Rapid Molecular Detection and Differentiation of Influenza Viruses A and B

Influenza is a contagious respiratory illness caused by influenza viruses A and B in humans and causes a significant amount of morbidity and mortality every year. The Influenza A and B assay was the first CLIA-waived molecular rapid flu test available. The Influenza A and B test works by employing isothermal amplification with influenza-specific primers followed by target detection with molecular beacon probes. Here, the performance of the Influenza A and B assay on frozen, archived nasopharyngeal swab (NPS) specimens stored in viral transport medium (VTM) were compared to a respiratory panel assay.
The performance of the Influenza A and B assay was evaluated by comparing the results to the respiratory panel reference method. The sensitivity for total influenza virus A was 67.5% (95% CI (CI), 56.6-78.5) and the specificity was 86.9% (CI, 71.0-100). For influenza virus B testing, the sensitivity and specificity were 90.2% (CI, 68.5-100) and 98.8% (CI, 68.5-100), respectively.
This system has the advantage of a significantly shorter test time than any other currently available molecular assay and the simple, pipette-free procedure runs on a fully integrated, closed, small-footprint system. Overall, the Influenza A and B assay evaluated in this study has the potential to serve as a point-of-care rapid influenza diagnostic test.

We describe a rapid, molecular-based Influenza A and B assay. The Influenza assay detects each target within 15 min by employing isothermal amplification with influenza-specific primers followed by target detection with molecular beacon probes. The Influenza A and B assay is user-friendly and required minimal hands-on time to perform.

High-throughput Detection of Respiratory Pathogens in Animal Specimens by Nanoscale PCR

Nanoliter scale real-time PCR uses spatial multiplexing to allow multiple assays to be run in parallel on a single plate without the typical drawbacks of combining reactions together. We designed and evaluated a panel based on this principle to rapidly identify the presence of common disease agents in dogs and horses with acute respiratory illness. This manuscript describes a nanoscale diagnostic PCR workflow for sample preparation, amplification, and analysis of target pathogen sequences, focusing on procedures that are different from microliter scale reactions. In the respiratory panel presented, 18 assays were each set up in triplicate, accommodating up to 48 samples per plate. A universal extraction and pre-amplification workflow was optimized for high-throughput sample preparation to accommodate multiple matrices and DNA and RNA based pathogens. Representative data are presented for one RNA target (influenza A matrix) and one DNA target (equine herpesvirus 1). The ability to quickly and accurately test for a comprehensive, syndrome-based group of pathogens is a valuable tool for improving efficiency and ergonomics of diagnostic testing and for acute respiratory disease diagnosis and management.

A High-throughput Platform for the Screening of Salmonella spp./Shigella spp.

Fecal-oral transmission of acute gastroenteritis occurs from time to time, especially when people who handled food and water are infected by Salmonella spp./Shigella spp. The gold standard method for the detection of Salmonella spp./Shigella spp. is direct culture but this is labor-intensive and time-consuming. Here, we describe a high-throughput platform for Salmonella spp./Shigella spp. screening, using real-time polymerase chain reaction (PCR) combined with guided culture. There are two major stages: real-time PCR and the guided culture. For the first stage (real-time PCR), we explain each step of the method: sample collection, pre-enrichment, DNA extraction and real-time PCR. If the real-time PCR result is positive, then the second stage (guided culture) is performed: selective culture, biochemical identification and serological characterization. We also illustrate representative results generated from it. The protocol described here would be a valuable platform for the rapid, specific, sensitive and high-throughput screening of Salmonella spp./Shigella spp.

Detection of Low Copy Number Integrated Viral DNA Formed by In Vitro Hepatitis B Infection

Hepatitis B Virus (HBV) is a common blood-borne pathogen causing liver cancer and liver cirrhosis resulting from chronic infection. The virus generally replicates through an episomal DNA intermediate; however, integration of HBV DNA fragments into the host genome has been observed, even though this form is not necessary for viral replication. The exact purpose, timing, and mechanism(s) by which HBV DNA integration occurs is not yet clear, but recent data show that it occurs very early after infection. Here, the in vitro generation and detection of HBV DNA integrations are described in detail. Our protocol specifically amplifies single copies of virus-cell DNA integrations and allows both absolute quantification and single-base pair resolution of the junction sequence. This technique has been applied to various HBV-susceptible cell types (including primary human hepatocytes), to various HBV mutants, and in conjunction with various drug exposures. We foresee this technique becoming a key assay to determine the underlying molecular mechanisms of this clinically relevant phenomenon.

Human Serum Anti-aquaporin-4 Immunoglobulin G Detection by Cell-based Assay

Anti-aquaporin-4 (AQP4) immunoglobulin G (IgG) is the core diagnostic biomarker for neuromyelitis optica spectrum disorders (NMOSD). The cell-based assay (CBA) is a widely used method to detect anti-AQP4 IgG in human serum with high sensitivity and specificity. Briefly, serum anti-AQP4 IgG is captured by AQP4-transfected cell that is fixed on the biochip then detected by a fluorescein-labelled secondary antibody. Fluorescence microscopy is utilized to visualize the fluorescence, and the intensity of fluorescence is evaluated by at least two experienced clinicians. A final diagnosis of NMOSD can be made based on the combination of anti-AQP4 IgG detection results, clinical manifestations, and neuroradiological findings. According to previous studies, CBA is more sensitive and specific than other anti-AQP4 IgG detection methods, and it can be applied to both clinical diagnosis and studies of NMOSD. The method has limitations; for example, an international scale to evaluate serum anti-AQP4 IgG titers is still lacking. Here, a detailed protocol for human serum anti-AQP4 IgG detection using CBA is described.

Detection of Extravascular Trypanosoma Parasites by Fine Needle Aspiration

Fine Needle Aspiration (FNA) is a routine diagnostic procedure essential to both medical and veterinary practices. It consists of the percutaneous aspiration of cells and/or microorganisms from palpable masses, organs or effusions (fluid accumulation in a body cavity) using a thin needle similar to the regular needle used for the venous puncture. The material collected by FNA is in general highly cellular, and the retrieved aspirate is then smeared, air dried, wet-fixed, stained and observed under a microscope. In the clinical context, FNA is an important diagnostic tool that serves as a guide to the appropriate therapeutic management. Because it is simple, fast, minimally invasive and requires limited investment in the laboratory and human resources, it is extensively used by veterinary practitioners, mostly in domestic, but also in farm animals. In studies using animal models, FNA has the advantage that it can be performed repeatedly in the same animal, enabling longitudinal studies through the collection of cells from tumors and organs/tissues over the course of the disease. In addition to routine microscopy, retrieved material can also be used for immunocytochemistry, electron microscopy, biochemical analysis, flow cytometry, molecular biology or in vitro assays. FNA has been used to identify the protozoan parasite Trypanosoma brucei in the gonads of infected mice, opening the possibility for a future diagnosis in cattle.
