1. マイクロバブルの社内準備
<ol><li>1,2-ジステアロイル-sn-グリセロ-3-ホスホコリンと1,2-ジステアロイル-sn-グリセロ-3-ホスホエタノールアミン-N-[アミノ(ポリエチレングリコール)-2000](アンモニウム塩)のモル比を9:1で計量する。マイクロバブル溶液1mLあたり0.5mgの脂質混合物が必要である。あるいは、脂質はクロロホルムで既に購入することができ、予め溶解した脂質を使用する場合はステップ1.3に進む。</li>
	<li>ガラスビーカー内の少量のクロロホルムに脂質を溶解する。</li>
	<li>クロロホルムをエバポレーターまたは窒素気流で蒸発させる。</li>
	<li>乾燥した脂質膜を、0.22 μm フィルターでろ過した PBS + 10% グリセロール + 10% プロピレングリコール溶液 10 mL で再水和します。</li>
	<li>再水和脂質溶液を55°C(脂質の溶融温度以上)に設定したウォーターバス超音波処理機に入れ、完全に溶解するまで超音波処理する。</li>
	<li>脂質溶液をオートクレーブ処理した1.5mL HPLCバイアルにアリコートし、セプタキャップにねじ込む。</li>
	<li>27G針を備えた5mLシリンジでバイアル内のすべての空気を吸引し、バイアル内に真空を作ります。</li>
	<li>付属のシリンジでオクトフルオロプロパンをバイアルに加え、キャニスターからガスを吸い込みます。シリンジ内の体積を読み取ることによって、バイアルに1〜2mLのオクタフルオロプロパンを充填する。</li>
	<li>各バイアルをパラフィンフィルムで密封し、冷蔵する。</li>
	<li>実験当日、バイアルを室温にし、0.5mLの0.9%NaCl溶液をバイアルに加え、バイアルをアマルガメーターに入れ、45秒(予め設定された時間)攪拌してマイクロバブルを生成させた。</li>
</ol>2. コールターカウンターによるマイクロバブル品質管理
<ol><li>マイクロバブル溶液をアマルガメータから取り出し、セプタを19G針で穿刺してバイアルからガスを排出する。</li>
	<li>19G針付き1mlシリンジを用いて5mLの濾過流動溶液にマイクロバブル溶液100μLを加えて2段階1:5,000段階希釈を行いマイクロバブル溶液を希釈し、次いで1:50希釈マイクロバブル溶液100μLを採取し、ピペットを用いてキュベット内の10mL濾過流動溶液にピペッティングする。</li>
	<li>電解質タンクに十分なフロー溶液があり、廃棄物タンクが空であることを確認してください。</li>
	<li>キュベットをコールターカウンタープラットフォームに置き、所定の位置にロックします。サンプル取得には30μmのアパーチャを使用してください。</li>
	<li>ソフトウェアで、標準操作方法(SOM)をロードし、[ SOM|濃度の編集 ]を選択します。5000倍の希釈率を入力します。</li>
	<li>ソフトウェアで、適切なファイル名を選択します。たとえば、microbubble_1_dateのようにします。</li>
	<li>キュベットをプラットフォームにロードして固定します。</li>
	<li>ソフトウェアで [ 実行] を選択|サンプル 濃度が10%未満であることをプレビューして確認します。この数値が10%より高い場合、より高い希釈倍率でマイクロバブルの新たな希釈を行う。</li>
	<li>「開始」を選択してサンプルの集録を開始し、初期読み出しを取得します。</li>
	<li>各測定後にろ過されたフロー溶液でコールターカウンターの開口部をすすぎます。手順2.2~2.9を繰り返して、3回の反復を取得します。</li>
	<li>超音波処理器の水浴に希釈マイクロバブル溶液とキュベットを置き、30秒間超音波処理します。</li>
	<li>超音波処理されたマイクロバブルで溶液を測定し、ブランクとしてラベルを付けます。</li>
	<li>ソフトウェアで、最初の読み出しから最終的な読み出しを減算します。これにより、マイクロバブルではなく、気体を含まない粒子が差し引かれます。</li>
	<li>ソフトウェアで 結果の表示 を選択し、マイクロバブル濃度、サイズ分布、平均サイズ、体積濃度を表示します。</li>
</ol>3. 蛍光抗体標識
<ol><li>マウス IgG を無添加で PBS 中に 1 mg/mL 溶液で入手します。</li>
	<li>キットにある製造元の指示に従って、マウスIgGを0.1 M重曹緩衝液中のAlexaFluor 647で1 mgのラベル付けします。この量の蛍光標識IgGは、5mg/kg用量の抗体を投与する5〜7匹の成体マウスでこの手順を実行するのに十分である。</li>
	<li>色素を0.1 M重曹緩衝液中のIgGの溶液に加え、室温で15分間インキュベートする。</li>
	<li>蛍光標識抗体を精製するには、抗体溶液をスピンカラムにピペッティングし、1,000 x g で5分間遠心分離します。遊離染料はカラムベッドに残ります。</li>
	<li>分光光度計を使用してタンパク質濃度を測定します。280nmおよび650nmにおけるコンジュゲート溶液の吸光度を測定する(A280およびA650)。次の式を使用して、サンプル中のタンパク質の濃度を計算します。 タンパク質濃度(M)=[A280-(A650 x 0.03)]×希釈倍率/203,000。</li>
	<li>分光光度計を使用して、次の式を使用してラベリングの程度を計算します。 モルタンパク質当たりの色素モル数=A650 x希釈倍率/239 000 xタンパク質濃度(M)。 注:許容可能な標識の程度は、タンパク質1モル当たり3〜7モルの色素であり、典型的には得られる標識の程度は約6である。</li>
</ol>4. 超音波のセットアップ
<ol><li>集束超音波システムを使用して、5mmスペーサーを水ボーラスに追加して、超音波焦点を水ボーラスの底から9mm下方に配置します。</li>
	<li>インライン脱気装置で20分間脱気した約300mLの脱イオン水で水ボーラスを満たします(酸素含有量は3ppm未満でなければなりません)。環状アレイを充填された水ボーラスに入れ、歯科用ミラーを使用して表面に気泡がないことを確認します。表面に存在する場合は、環状アレイを取り外し、水中ボーラスに入れ替えます。</li>
	<li>アプリケーションソフトウェアを起動します。波形メニューで、「 波形デューティサイクルを設定」を選択します。設定はPRF(Hz)10、デューティサイクル10%、フォーカス80mm、中心周波数1MHz、振幅(MPaピーク負圧)0.65MPa、メカニカルインデックス=0.65です。 Set キーを押して波形を定義し、メモリに保存します。 注:集束超音波システムは、校正されたハイドロフォンによって行われた測定値から製造業者によって事前に較正されています。</li>
	<li>集束超音波システムソフトウェアで、治療計画を定義します。これには、複数の個々の治療部位からなる治療領域を定義し、それらの治療部位のそれぞれでとるべきアクションを定義することが必要である。この場合、治療ゾーンはマウス脳の1半球である。</li>
	<li>モーションコントローラウィンドウで、[ スキャン ]タブに移動し、x次元のモーションの開始値、停止値、インクリメント値を入力し、y方向の動きの開始値、停止値、インクリメント値 を入力します 。X: 開始 -4、停止 3.50、Y: -3.00、停止 3、インクリメント 1.5、# ループ: 1 の値を入力します。</li>
	<li>治療部位のアクションを定義します。モーションコントローラウィンドウで、[ イベント ]ボタンを選択します。 スクリプト編集ウィンドウで、各治療部位で選択した順序で実行されるアクションのリストを選択します。スクリプトウィンドウの上部にある [移動タイプ ]をラスターグリッドに設定します。「イベント」タブで 、「アクションの追加」 を選択してスクリプト・パネルに移動し、「 同期的に移動」、「トリガー arb を 開始」、「 待機」、「トリガー arb を停止」 を追加します。待機アクションをクリックし、6, 000 ミリ秒の待機時間を選択します。 注:これらの設定により、治療は1.5 mm間隔で6 x 5グリッドの処理スポットになり、各スポットの治療期間は6秒になります。マウスの脳を超音波処理する総持続時間は約3分です。このサイズの治療グリッドは、体重約30gの成体C57/Bl6マウスに適しています。治療スポットのグリッドのサイズは、マウスのサイズに応じて上下に調整することができます。</li>
</ol>5. 動物の準備
<ol><li>0.1gの正確な天秤でマウスの重量を量ります。</li>
	<li>マウスを腹腔内に90mg / kgケタミンおよび6mg / kgキシラジンで麻酔する。つま先のピンチで反射神経がないかテストします。あるいは、マウスは、適切なフェイスマスクを有する適切な吸入麻酔装置を用いてイソフルランで麻酔することができる。イソフルランを使用する場合は、低体温症を防ぐために超音波検査中にマウスをヒートパッドに置く必要があります。</li>
	<li>電気カミソリを使用して動物の頭から髪を剃り、綿芽で脱毛クリームを塗布し、2〜3分間、または湿ったガーゼで髪をきれいに拭き取るまで放置します。脱毛クリームがマウスの目に入り込まないように注意してください。</li>
	<li>マウスの頭の中心に永久マーカーを付けます。トランスデューサの中心には穴があり、トランスデューサの焦点と焦点を視覚的に位置合わせすることができます。</li>
	<li>以前に底部が切断され、超音波ゲルで計量ボートの底に接着されたプラスチックラップと交換された小さな計量ボートを充填する。これは8mmのスペーサーとして機能し、マウスのヘッドに良好な結合を提供し、マウスのヘッドに合わせた探触子の焦点の目視検査を可能にします。</li>
</ol>6. マイクロバブル調製
<ol><li>マイクロバブルバイアルを室温まで温める。活性化するには、0.5mLの0.9%NaCl溶液をバイアルに加え、アマルガメータに入れて45秒間攪拌してマイクロバブルを生成する。</li>
	<li>セプタを27G針で突き刺してバイアルを通気します。</li>
</ol>7. 超音波治療
<ol><li>マイクロバブルのバイアルを反転させ、1μL/gの体重の溶液を静かに引き出す。これに蛍光標識抗体の溶液を加え、シリンジ内で穏やかに混合する。注入される最大容量は150μLである。 注:社内で調製されたマイクロバブルは、臨床的に使用されているマイクロバブル(例えば、Definityマイクロバブル)よりも約60倍濃度が低い。注入されるマイクロバブルの数が臨床的に使用されているものと類似するように体積または濃度を調整する(すなわち、マイクロバブルの数が臨床的に使用されているものと同様である(すなわち、マイクロバブルの数が臨床的に使用されているものと同様である。定義1.2 x 108 マイクロバブル/ kg体重)。</li>
	<li>マイクロバブルと抗体溶液を眼窩下へ注入し、穏やかでゆっくりと注入するように注意する。次に、マウスの目に眼科用軟膏を塗布する。</li>
	<li>マウスをヘッドホルダー(材料表を参照)に置き、マウスの鼻を固定します。次に、超音波ゲルで満たされた小さな計量ボートを頭の上に置きます。</li>
	<li>計量ボートの超音波ゲルの上に座るまで、水ボーラスを下げます。</li>

	<li>[スキャンの完了] を選択します。手順 7.3 ~ 7.6 の所要時間は 2 分です。</li>
	<li>一貫性を保つために、マイクロバブルの注入から完全スキャンの選択までの間に2分の遅延があるようにタイマーを設定します。</li>
	<li>治療が完了したら、眼科用軟膏を眼に塗布し、マウスを温めた回収チャンバに入れる。処置中に低体温が観察された場合、処置中に補助的な熱を提供するために、加温パッドをマウスの下に置くことができる。</li>
</ol>8. 組織の採取と加工
<ol><li>超音波送達後の関心のある時点(脳内の高レベルの抗体を検出するために少なくとも1時間)に、ペントバルビトン溶液(100mg / kg)の過剰摂取でマウスを深く麻酔し、30mLのPBSでマウスを経心線灌流する。良好な灌流は、脳に送達された蛍光標識抗体を特異的に検出するために必要とされる。</li>
	<li>脳を集め、4%PFAに4°Cで24時間浸漬して固定し、PBSで洗浄する。</li>
	<li>脳をトレイの上に置き、700nmチャンネルの画像を取得することによって、赤外線スキャナで脳を画像化します。 注:固定後、脳をPFAから除去し、PBSで洗浄し、切片化する。あるいは、エチレングリコール凍結保護剤溶液中に4°Cで24時間、または水没するまで入れ、次いで新しい凍結保護剤含有バイアルに移動し、長期保存のために−20°Cで置くことができる。</li>
	<li>ビブラートームを用いてPBSで脳風邪を切開する。脳をプラットフォームに接着し、30〜40μmの切片を切断してPBSに集める。 注:リソソーム自己蛍光(約12ヶ月以上経過した動物からの切片で流行)は、細菌の増殖をブロックするために、室温およびアジド(0.02%)を含むPBS中で、軽いチャンバーボックス内の切片の一晩照明によって漂白されるべきである。</li>
</ol>9. 組織染色と画像取得
<ol><li>切片をブロッキング溶液(0.2%トリトン/PBS中の5%BSA)に室温で2時間移送し、次いで溶液を0.2%トリトン/PBSで置換して切片3xを洗浄する。</li>
	<li>切片を、Iba1(希釈1:1,000)およびCD68(希釈1:500)に対する一次抗体と共に、0.2%Triton/PBS中で一晩4°Cでインキュベートし、続いて0.2%Triton/PBSで3回洗浄する。</li>
	<li>切片を蛍光二次抗体(希釈1:500)で室温で2時間インキュベートし、続いて0.2%Triton/PBSのみで3回洗浄します。 注:核は、DAPI溶液(0.5μg/mL)を使用して染色することもできます。</li>
	<li>切片をスライドとカバースリップに、ハードセットの取り付けメディアで移します。可視化と画像取得の前に(一晩)取り付け媒体が固化するのに十分な時間を確保してください。</li>
	<li>共焦点顕微鏡を用いて、共焦点顕微鏡および少なくとも40倍の倍率の対物レンズを用いて、超音波標的脳領域のzスタック画像(少なくとも10μm深さおよび0.3μmステップサイズ、動物あたり10画像)を得る。 メモ:信号ダイナミックレンジ内の画像を取得するように注意し、露出不足/露出過多を避けてください。顕微鏡で使用される対応するソフトウェア形式で画像ファイルを保存します。</li>
</ol>10. 画像解析
<ol><li>画像解析ソフトウェアのファイルインポーターを使用して、zスタック顕微鏡ファイルを変換します。</li>
	<li>変換したファイルをソフトウェアに開き、Iba1チャネルの強度を調整してミクログリア細胞を観察します。</li>
	<li>単一のミクログリアセルの周りにボックスを描画してトリミングし、「トリミング」を選択して新しいファイルを保存します。</li>

MRI-guided Disruption of the Blood-brain Barrier using Transcranial Focused Ultrasound in a Rat Model

Focused ultrasound (FUS) disruption of the blood-brain barrier (BBB) is an increasingly investigated technique for circumventing the BBB1-5. The BBB is a significant obstacle to pharmaceutical treatments of brain disorders as it limits the passage of molecules from the vasculature into the brain tissue to molecules less than approximately 500 Da in size6. FUS induced BBB disruption (BBBD) is temporary and reversible4 and has an advantage over chemical means of inducing BBBD by being highly localized. FUS induced BBBD provides a means for investigating the effects of a wide range of therapeutic agents on the brain, which would not otherwise be deliverable to the tissue in sufficient concentration. While a wide range of ultrasound parameters have proven successful at disrupting the BBB2,5,7, there are several critical steps in the experimental procedure to ensure successful disruption with accurate targeting. This protocol outlines how to achieve MRI-guided FUS induced BBBD in a rat model, with a focus on the critical animal preparation and microbubble handling steps of the experiment.

Real-Time Intravital Multiphoton Microscopy to Visualize Focused Ultrasound and Microbubble Treatments to Increase Blood-Brain Barrier Permeability

The blood-brain barrier (BBB) is a key challenge for the successful delivery of drugs to the brain. Ultrasound exposure in the presence of microbubbles has emerged as an effective method to transiently and locally increase the permeability of the BBB, facilitating para- and transcellular transport of drugs across the BBB. Imaging the vasculature during ultrasound-microbubble treatment will provide valuable and novel insights on the mechanisms and dynamics of ultrasound-microbubble treatments in the brain.
Here, we present an experimental procedure for intravital multiphoton microscopy using a cranial window aligned with a ring transducer and a 20x objective lens. This set-up enables high spatial and temporal resolution imaging of the brain during ultrasound-microbubble treatments. Optical access to the brain is obtained via an open-skull cranial window. Briefly, a 3-4 mm diameter piece of the skull is removed, and the exposed area of the brain is sealed with a glass coverslip. A 0.82 MHz ring transducer, which is attached to a second glass coverslip, is mounted on top. Agarose (1% w/v) is used between the coverslip of the transducer and the coverslip covering the cranial window to prevent air bubbles, which impede ultrasound propagation. When sterile surgery procedures and anti-inflammatory measures are taken, ultrasound-microbubble treatments and imaging sessions can be performed repeatedly over several weeks. Fluorescent dextran conjugates are injected intravenously to visualize the vasculature and quantify ultrasound-microbubble induced effects (e.g., leakage kinetics, vascular changes). This paper describes the cranial window placement, ring transducer placement, imaging procedure, common troubleshooting steps, as well as advantages and limitations of the method.

This protocol describes the surgical and technical procedures that enable real-time in vivo multiphoton fluorescence imaging of the rodent brain during focused ultrasound and microbubble treatments to increase blood-brain barrier permeability.

Delivery of Antibodies into the Murine Brain via Convection-enhanced Delivery

Convection-enhanced delivery (CED) is a neurosurgical technique enabling effective perfusion of large brain volumes using a catheter system. Such an approach provides a safe delivery method by-passing the blood brain barrier (BBB), thus allowing treatment with therapeutics with poor BBB-permeability or those for which systemic exposure is not desired, e.g., due to toxicity. CED requires optimization of the catheter design, injection protocol, and properties of the infusate. With this protocol we describe how to perform CED of a solution containing up to 20 &#181;g of an antibody into the caudate putamen of mice. It describes preparation of step catheters, testing them in vitro and performing the CED in mice using a ramping injection program. The protocol can be readily adjusted for other infusion volumes and can be used for injecting various tracers or pharmacologically active or inactive substances, including chemotherapeutics, cytokines, viral particles, and liposomes.

A Benchtop Approach to the Location Specific Blood Brain Barrier Opening using Focused Ultrasound in a Rat Model

Stereotaxic surgery is the gold standard for localized drug and gene delivery to the rodent brain. This technique has many advantages over systemic delivery including precise localization to a target brain region and reduction of off target side effects. However, stereotaxic surgery is highly invasive which limits its translational efficacy, requires long recovery times, and provides challenges when targeting multiple brain regions. Focused ultrasound (FUS) can be used in combination with circulating microbubbles to transiently open the blood brain barrier (BBB) in millimeter sized regions. This allows intracranial localization of systemically delivered agents that cannot normally cross the BBB. This technique provides a noninvasive alternative to stereotaxic surgery. However, to date this technique has yet to be widely adopted in neuroscience laboratories due to the limited access to equipment and standardized methods. The overall goal of this protocol is to provide a benchtop approach to FUS BBB opening (BBBO) that is affordable and reproducible and can therefore be easily adopted by any laboratory.

A High-Throughput Image-Guided Stereotactic Neuronavigation and Focused Ultrasound System for Blood-Brain Barrier Opening in Rodents

The blood-brain barrier (BBB) has been a major hurdle for the treatment of various brain diseases. Endothelial cells, connected by tight junctions, form a physiological barrier preventing large molecules (&#62;500 Da) from entering the brain tissue. Microbubble-mediated focused ultrasound (FUS) can be used to induce a transient local BBB opening, allowing larger drugs to enter the brain parenchyma.
In addition to large-scale clinical devices for clinical translation, preclinical research for therapy response assessment of drug candidates requires dedicated small animal ultrasound setups for targeted BBB opening. Preferably, these systems allow high-throughput workflows with both high-spatial precision as well as integrated cavitation monitoring, while still being cost effective in both initial investment and running costs.
Here, we present a bioluminescence and X-ray guided stereotactic small animal FUS system that is based on commercially available components and fulfills the aforementioned requirements. A particular emphasis has been placed on a high degree of automation facilitating the challenges typically encountered in high-volume preclinical drug evaluation studies. Examples of these challenges are the need for standardization in order to ensure data reproducibility, reduce intra-group variability, reduce sample size and thus comply with ethical requirements and decrease unnecessary workload. The proposed BBB system has been validated in the scope of BBB opening facilitated drug delivery trials on patient-derived xenograft models of glioblastoma multiforme and diffuse midline glioma.

Focused Ultrasound Induced Blood-Brain Barrier Opening for Targeting Brain Structures and Evaluating Chemogenetic Neuromodulation

Acoustically Targeted Chemogenetics (ATAC) allows for the noninvasive control of specific neural circuits. ATAC achieves such control through a combination of focused ultrasound (FUS) induced blood-brain barrier opening (FUS-BBBO), gene delivery with adeno-associated viral (AAV) vectors, and activation of cellular signaling with engineered, chemogenetic, protein receptors and their cognate ligands. With ATAC, it is possible to transduce both large and small brain regions with millimeter precision using a single noninvasive ultrasound application. This transduction can later allow for a long-term, noninvasive, device-free neuromodulation in freely moving animals using a drug. Since FUS-BBBO, AAVs, and chemogenetics have been used in multiple animals, ATAC should also be scalable for the use in other animal species. This paper expands upon a previously published protocol and outlines how to optimize the gene delivery with FUS-BBBO to small brain regions with MRI-guidance but without a need for a complicated MRI-compatible FUS device. The protocol, also, describes the design of mouse targeting and restraint components that can be 3D-printed by any lab and can be easily modified for different species or custom equipment. To aid reproducibility, the protocol describes in detail how the microbubbles, AAVs, and venipuncture were used in ATAC development. Finally, an example data is shown to guide the preliminary investigations of studies utilizing ATAC.

Monitoring Blood-Brain Barrier Opening in Rats with a Preclinical Focused Ultrasound System

The brain has a highly selective semipermeable blood barrier, termed the blood-brain barrier (BBB), which prevents the delivery of therapeutic macromolecular agents to the brain. The integration of MR-guided low-intensity pulsed focused ultrasound (FUS) with microbubble pre-injection is a promising technique for non-invasive and non-toxic BBB modulation. MRI can offer superior soft-tissue contrast and various quantitative assessments, such as vascular permeability, perfusion, and the spatial-temporal distribution of MRI contrast agents. Notably, contrast-enhanced MRI techniques with gadolinium-based MR contrast agents have been shown to be the gold standard for detecting BBB openings. This study outlines a comprehensive methodology involving MRI protocols and animal procedures for monitoring BBB opening in a rat model. The rat model provides the added benefit of jugular vein catheter utilization, which facilitates rapid medication administration. A stereotactic-guided preclinical FUS transducer facilitates the refinement and streamlining of animal procedures and MRI protocols. The resulting methods are characterized by reproducibility and simplicity, eliminating the need for specialized surgical expertise. This research endeavors to contribute to the optimization of preclinical procedures with rat models and encourage further investigation into the modulation of the BBB to enhance therapeutic interventions in neurological disorders.

The skull of an anesthetized mouse is exposed, and a burr hole is created at the target area for antibody delivery. A step catheter containing antibodies is inserted at the target site, and via an infusion pump, a pressure gradient is created to deliver antibodies at a controlled rate.
