1. mESC-Kultur
<ol><li>Beschichten Sie eine 10 cm gewebekulturbehandelte Platte mit 0,1% Gelatine und lassen Sie die Gelatine mindestens 15-30 min aushärten, bevor Sie sie aussaugen.</li>
	<li>Samen γ-bestrahlten embryonalen Fibroblasten (MEFs) der Maus einen Tag vor der Kultivierung der mESCs im vorgewärmten mESC-Medium (Dulbeccos modifiziertes Eagle-Medium (DMEM) mit 15% fetalem Rinderserum (FBS), nicht-essentiellen Aminosäuren, β-Mercaptoethanol, L-Glutamin, Penicillin/Streptomycin, Natriumpyruvat, LIF, PD0325901 (PD) und Chir99021 (CH)).</li>
	<li>Lassen Sie die γ bestrahlten MEFs sich absetzen und an der Plattenoberfläche befestigen, bevor Sie E14-Zellen kultivieren.</li>
	<li>E14 ESCs im 37 °C Wasserbad auftauen und die Zellen in einem 15 cm konischen Röhrchen mit warmem mESC-Medium schnell übertragen. Die Zellen bei 200 x g für 3 min pelletieren und den Überstand entfernen.</li>
	<li>Resuspendieren Sie die Zellen in 10 ml mESC-Medium und platten Sie die Zellen auf der Kulturplatte, die die zuvor γ bestrahlten MEFs enthält. Inkubieren Sie die Zellkultur in einem 37 °C Inkubator unter 5%CO2.</li>
	<li>Für die Kulturübergabe das Medium ansaugen und die Platte mit sterilen 1x PBS waschen. Fügen Sie genügend 0,05% Trypsin hinzu, um die Plattenoberfläche zu bedecken, und inkubieren Sie bei 37 °C für 3 min.</li>
	<li>Neutralisieren Sie das Trypsin mit mESC-Medium und Pipette, um eine Einzelzellsuspension zu erzeugen. Zentrifugieren Sie die Zellen bei 200 x g für 3 min und entfernen Sie den Überstand.</li>
	<li>Zählen Sie die Zellen mit einem Hämozytometer oder Zellzähler und säen Sie etwa 5,0 x 105 Zellen in eine 10 cm große Kulturplatte.</li>
	<li>Resuspendieren Sie die Zellen in 10 ml mESC-Medium, platten Sie die Zellen auf die mit Gelatine beschichtete Gewebekulturplatte und inkubieren Sie Kulturen wie zuvor beschrieben. HINWEIS: Es wird empfohlen, dass mESCs alle 2 Tage durchquert werden, um zu verhindern, dass sich die Zellen in ihren Kolonien differenzieren. Phenolrot im Medium fungiert ausschließlich als pH-Indikator und kann je nach Zelldichte früher als 2 Tage gelblich (saurer) werden. Daher kann es notwendig sein, das Medium jeden Tag zu wechseln. Die γ-bestrahlten MEFs werden schließlich nach ein paar Passagen absterben.</li>
</ol>2. EB-, NPC- und Neuronendifferenzierung
<ol><li>Führen Sie das bereits erwähnte Kultur-Passaging-Protokoll durch und zählen Sie die Zellen (Schritte 1.7–1.10).</li>
	<li>Hängende Tropfenmethode (Tag 0)<ol>
<li>Zählen Sie für eine 10 cm Große Zellkulturplatte etwa 2,5 x 104 Zellen, wobei 5,0 x10 2 Zellen in einem 20 μL-Differenzierungsmedium suspendiert werden (DMEM mit 15% FBS, nicht-essentiellen Aminosäuren, β-Mercaptoethanol, L-Glutamin, Penicillin/Streptomycin und Natriumpyruvat). Etwa fünfzig 20 μL Tröpfchen, die die Zellen enthalten, können auf einer 10 cm großen Platte plattiert werden.</li>
		<li>Aliquotieren Sie die entsprechende Anzahl von Zellen und zentrifugieren Sie dann die Zellen bei 200 x g für 3 min und entfernen Sie den Überstand.</li>
		<li>Resuspendieren Sie die Zellen im geeigneten Volumen des Differenzierungsmediums für eine Zelldichte von 5,0 x10 2 Zellen pro 20 μL (z. B. 2,5 x 104 Zellen in 1 ml Differenzierungsmedium).</li>
		<li>Mit einer Mikropipette oder einer Repeaterpipette 20 μL Tröpfchen der Zellsuspension auf den Deckel der Gewebekulturplatte geben. Stellen Sie sicher, dass die Tröpfchen nicht zu nahe beieinander liegen, um zu verhindern, dass sie verschmelzen. HINWEIS: Die Tröpfchen können entweder auf einer mit Gewebekulturen behandelten Befestigungsplatte oder einer Suspensionsplatte plattiert werden, da sie auf dem Deckel und nicht auf der Platte selbst platziert werden. Für einen praktikableren und sterileren Ansatz plätten Sie die Tröpfchen auf einer Befestigungs-Gewebekulturplatte und übertragen Sie sie wie unten beschrieben auf eine Suspensionsplatte.</li>
		<li>Füllen Sie die Platte mit 5–10 ml 1x PBS auf und setzen Sie den Deckel vorsichtig wieder auf die Platte. Inkubieren Sie die Kultur im 37 °C Inkubator. HINWEIS: PBS wird der Kulturplatte zugesetzt, um zu verhindern, dass die Tröpfchen austrocknen.</li>
	</ol></li>
	<li>Am Tag 2sammeln Sie mit einer Mikropipette die Tröpfchen aus dem Deckel und legen Sie sie in eine 10 cm große Zellkultur-Suspensionsplatte, die mit 10 ml Differenzierungsmedium gefüllt ist. Inkubieren Sie die Kultur auf einem Orbitalschüttler, der mit niedriger Geschwindigkeit im Inkubator schüttelt.</li>
	<li>Am Tag 4,um die EBs zu ernten, sammeln Sie die Zellen, zentrifugieren Sie bei 200 x g für 3 min und entfernen Sie den Überstand. HINWEIS: Die EBs können auch mit 1x PBS gemäß den Anforderungen nachfolgender Experimente gewaschen werden.</li>
	<li>Um das Verfahren fortzusetzen und die EB-Differenzierung in neuronale Vorläuferzellen (NPCs) zu induzieren, bereiten Sie das Differenzierungsmedium mit 5 μM Retinsäure (RA) vor.</li>
	<li>Entfernen Sie das alte Medium, indem Sie die EBs bei 100 x g für 3 min pelletieren oder lassen Sie die EBs absetzen, bevor Sie das alte Medium aussaugen. 10 mL des Differenzierungsmediums, das 5 μM RA enthält, in die Kulturplatte geben.</li>
	<li>Ersetzen Sie am Tag 6mindestens die Hälfte des Mediums durch ein frisches Medium, das 5 μM RA enthält, indem Sie die Platte kippen und das Medium wie oben beschrieben pipettieren. HINWEIS: Es wird empfohlen, an den Tagen 5 und 7 mindestens die Hälfte des Mediums durch frisches RA-haltiges Medium zu ersetzen. Beachten Sie den phenolroten Indikator; Wenn es gelblich wird, ist es am besten, das gesamte Medium zu ersetzen.</li>
	<li>Am Tag 8erntet ihr die NPCs, indem ihr die Zellen sammelt, 3 min lang bei 200 x g zentrifugiert und den Überstand entfernt. HINWEIS: Die NPCs können auch mit 1x PBS gewaschen werden, je nach den Bedürfnissen nachfolgender Experimente. Bei Bedarf können NPCs eingefroren und für die spätere Kultur und Analyse wieder aufgetaut werden. Sollen die NPCs kultiviert werden, kann Accutase auch als Alternative zu Trypsin verwendet werden.</li>
	<li>Um das Verfahren fortzusetzen und NPCs zu Neuronen zu differenzieren, sammeln Sie NPCs in einem 15 ml konischen Rohr durch Zentrifugation, dissoziieren Sie sie mit Trypsin und inkubieren Sie sie bei 37 ° C für 3 min. Pipettieren Sie die NPCs, um sicherzustellen, dass alle NPC-Aggregate dissoziiert sind und neutralisieren Sie das Trypsin mit dem Medium.</li>
	<li>Filtern Sie die Zellen mit einem 40 μm Nylon-Zellsieb und zählen Sie die Zellen, bevor Sie sie mit einer Dichte von 1,5 x 105/ cm2 in N2-Medium (DMEM / F12-Medium + 3 mg / ml Glukose + 3 mg / ml lipidreiches Rinderserumalbumin (LBSA) + 1:100 N2-Präparat + 10 ng / ml bFGF + 50 U / ml Pen / Streptokokken + 1 mM L-Glutamin) auf einer mit Gewebekultur behandelten Platte für nachfolgende PCR- und Western-Blot-Experimente plattieren; oder auf einer Gewebekulturkammer für Immunfluoreszenzexperimente.</li>
	<li>Ersetzen Sie am Tag 9das alte Medium durch ein frisches N2-Medium.</li>
	<li>Wechseln Sie am Tag 10das N2-Medium mit N2/B27-Medium (50% DMEM/F12 und 50% neural basal, 3 mg/ml LBA, 1:200 N2-Präparat, 1:100 B27-Präparat, 50 U/ml Pen/Streptokokken und 1 mM L-Glutamin).</li>
	<li>An den Tagen 11-12ernten Sie die Neuronen wie folgt. Waschen Sie die Zellen mit 1x PBS, fügen Sie Trypsin hinzu und inkubieren Sie die Kultur im 37 ° C-Inkubator für 3 min, bevor Sie das Trypsin mit Medium neutralisieren und 3 min bei 200 x g zentrifugieren.</li>
</ol>3. Charakterisierung von mES-Zellen und differenzierten Zellen
<ol><li>Alkalischer Phosphatase (AP) Assay 
	<ol>
<li>Verwenden Sie ein Kit, um die alkalische Phosphataseaktivität zu beurteilen (siehe Materialtabelle).</li>
		<li>Entfernen Sie das Medium von der Kulturplatte und waschen Sie die ESCs mit 1x PBS.</li>
		<li>1 ml der fixen Lösung (besteht aus Formaldehyd und Methanol), die mit dem Kit geliefert wurde, auf die Platte geben und bei Raumtemperatur für 2–5 min inkubieren. HINWEIS: Eine Überinkubation in der Fixlösung kann die AP-Aktivität beeinträchtigen.</li>
		<li>Entfernen Sie die Fixlösung, waschen Sie die ESCs mit 1x PBS und lassen Sie eine gewisse Menge PBS in der Platte. HINWEIS: Halten Sie die ESCs in PBS feucht, um die AP-Aktivität nicht zu beeinträchtigen.</li>
		<li>Bereiten Sie die AP-Lösung vor, indem Sie die A-, B- und C-Substratlösungen im Verhältnis 1:1:1 mischen. Mischen Sie zuerst A- und B-Lösungen und inkubieren Sie die Mischung bei Raumtemperatur für 2 min, bevor Sie die C-Lösung hinzufügen.</li>
		<li>Entfernen Sie die 1x PBS, und fügen Sie die zuvor vorbereitete AP-Lösung hinzu.</li>
		<li>Inkubieren Sie die ESCs für ca. 15 min im Dunkeln, indem Sie die Kulturplatte mit Alufolie umwickeln oder Schritt 3.5 in einem dunklen Raum durchführen.</li>
		<li>Überwachen Sie die Reaktion und entfernen Sie die Reaktionslösung, wenn die Lösung hell wird, um unspezifische Flecken zu vermeiden.</li>
		<li>Waschen Sie die ESCs zweimal mit 1x PBS.</li>
		<li>Verhindern Sie das Austrocknen der Probe, indem Sie die ESCs mit 1x PBS oder Montagemedium abdecken. HINWEIS: Für den AP-Ausdruck erscheint ein roter oder violetter Fleck. Der Teller kann in einem 4 °C Kühlschrank aufbewahrt werden.</li>
	</ol></li>
	<li>RT-qPCR 
	<ol>
<li>Sammeln Sie Zellen in verschiedenen Stadien, indem Sie die Schritte 1.6-1.7 und 2.4 für ES-Zellen bzw. EBs bzw. NPCs ausführen.</li>
		<li>Isolieren Sie RNA mit RNA, DNA und Proteinextraktionslösung (siehe Tabelle der Materialien).</li>
		<li>Generieren Sie cDNA mit einem Reverse-Transkriptase-Kit (siehe Materialtabelle)und folgen Sie dem Handbuch des Herstellers.</li>
	</ol></li>
	<li>Fixierung und Einbettung 
	<ol>
<li>Ernten Sie EBs und NPCs wie oben beschrieben (Schritt 2.6) und fixieren Sie sie mit 4% Paraformaldehyd (PFA) Lösung in 1x PBS für 30 min bei Raumtemperatur.</li>
		<li>Entfernen Sie die PFA und waschen Sie die Probe mit 1x PBS für 5 min.</li>
		<li>Die Probe wird in eine serielle Verdünnung von 1x PBS-, 10%-, 20%- und 30%-Saccharoselösungen bei 25°u201228 °C gegeben, wobei die Probe nach 30 min Inkubation in die nächste Lösung überführt wird. HINWEIS: Die Probe kann in der 30%igen Saccharoselösung bei 4 °C gelagert werden, bevor mit dem Einbettungsschritt fortgefahren wird.</li>
		<li>Befeuchten Sie die Pipettenspitze mit Saccharoselösung, bevor Sie die Probe (ohne sie zu stapeln) in die Mitte der Kryoform geben und die überschüssige Flüssigkeit auspipetieren. HINWEIS: Filterpapier kann auch verwendet werden, um die überschüssige Lösung zu entfernen. Die Benetzung der Pipettenspitze mit Saccharoselösung ist wichtig, um zu verhindern, dass die EBs und NPCs an den Wänden der Spitze haften bleiben.</li>
		<li>Geben Sie der Form vorsichtig eine optimale Schneidtemperaturlösung (OCT) hinzu, ohne die Proben erneut zu verwenden, und entfernen Sie überschüssige Blasen mit einer Pipette.</li>
		<li>Legen Sie die Form mit der Probe bei niedriger Geschwindigkeit auf einen Labormischer, um die Probe 15 Minuten lang leicht zu rühren. Dies hilft, die EBs und NPCs unten zu platzieren, wenn sie in der OCT-Lösung resuspendiert werden.</li>
		<li>Frieren Sie die Probe schnell ein, indem Sie die Form in flüssigen Stickstoff oder auf Trockeneis legen. HINWEIS: Die Proben können in einem Gefrierschrank von -70 °C gelagert werden, bevor sie mit dem nächsten Schritt fortfahren.</li>
	</ol></li>
	<li>Kryosektion 
	<ol>
<li>Stellen Sie den Kryostaten auf -20 bis -18 °C ab, bevor Sie die Probe auf das Gerät übertragen.</li>
		<li>Lösen Sie den gefrorenen OCT-Block von der Form und befestigen Sie ihn mit einer kleinen OCT-Lösung auf der Oberfläche des Halters.</li>
		<li>Richten Sie den OAT-Block so aus, dass die EBs und NPCs dem Blade am nächsten sind, um sicherzustellen, dass die Probe während des Schneidens nicht verloren geht.</li>
		<li>Schneiden Sie vorsichtig 10 μm des Blocks und achten Sie genau auf die Scheiben, die die Probe enthalten.</li>
		<li>Legen Sie die OCT-Scheibe, die die Probe enthält, schnell auf den Glasträger mit Gewebeeinbettung und lassen Sie die OCT-Scheiben 1 h bei Raumtemperatur an der Luft trocknen. HINWEIS: Die Proben können bei -70 °C für die spätere Verwendung gelagert werden.</li>
	</ol></li>
	<li>Immunfluoreszenz (IF) 
	<ol>
<li>Blockieren Sie OCT-Abschnitte oder Kulturkammern, die Neuronen in 10% normalem Eselserum / 0,1% Triton X-100 in 1x PBS für 1 h bei Raumtemperatur enthalten.</li>
		<li>Inkubieren Sie die Proben in primären Antikörpern, verdünnt in 5% normalem Eselserum/0,05% Triton X-100 in 1x PBS über Nacht bei 4 °C.</li>
		<li>Proben in 1x PBS/0,1% Triton X-100 dreimal für 5 min pro Wäsche waschen.</li>
		<li>Inkubieren Sie die Proben mit sekundärem Antikörper, verdünnt in 5% normalem Eselserum/0,05% Triton X-100 in 1x PBS für 1 h bei Raumtemperatur.</li>
		<li>Proben in 1x PBS/0,1% Triton X-100 dreimal für jeweils 5 min waschen und die Proben dann in 1 μg/ml DAPI inkubieren.</li>

	<li>RNA-seq-Analyse

<li>Sammeln Sie die Zellen in verschiedenen Stadien und führen Sie eine RNA-Extraktion durch (siehe Schritt 3.2).</li>

<li>Sammeln Sie ES-Zellen, indem Sie die Schritte 1.6-1.7 ausführen und die Zellen in Medium resuspendieren. Sammle die EBs und NPCs, indem du Schritt 2.4 befolgest, und resuspendiere die Zellen in Medium.</li>

Die Methode zur neuronalen Differenzierung von embryonalen Stammzellen der Maus ist seit Jahrzehnten etabliert, und die Forscher haben die vorherigen Protokolle weiter modifiziert oder neue für verschiedene Zwecke geschaffen7,10,11. Wir verwendeten eine Reihe von Assays, um die Effizienz und den Fortschritt der Differenzierungsstufen von mESCs zu Neuronen umfassend zu analysieren, die bei der Analyse anderer Abstammungsdifferen...

Seit ihrer ersten Ableitung aus der inneren Zellmasse der sich entwickelnden Mausblastozysten1,2, wurden embryonale Mausstammzellen (mESC) als leistungsfähige Werkzeuge zur Untersuchung der Selbsterneuerung und Differenzierung von Stammzellen verwendet3. Darüber hinaus führt die Untersuchung der mESC-Differenzierung zu einem enormen Verständnis molekularer Mechanismen, die die Effizienz und Sicherheit in der stammzellbasierten Therapie bei der Behandlung von Krankheiten wie neurodegenerativen Erkrankungen verbessern können4. Im Vergleich zu Tiermodellen bietet dieses In-vitro-System viele Vorteile, darunter Einfachheit in der Praxis und Bewertung, geringe Kosten bei der Pflege von Zelllinien im Gegensatz zu Tieren und relative Leichtigkeit bei genetischen Manipulationen. Die Effizienz und Qualität differenzierter Zelltypen wird jedoch häufig durch unterschiedliche Linien von mES-Zellen sowie die Differenzierungsmethoden5,6beeinflusst. Auch die traditionellen Assays zur Bewertung der Differenzierungseffizienz beruhen auf der qualitativen Untersuchung ausgewählter Markergene, denen es an Robustheit mangelt und die daher globale Veränderungen in der Genexpression nicht erfassen.
Hier wollen wir eine Batterie von Assays zur systematischen Beurteilung der neuronalen Differenzierung einsetzen. Mit traditionellen In-vitro-Analysen an ausgewählten Markern und RNA-seq etablieren wir eine Plattform zur Messung der Differenzierungseffizienz sowie der transkriptomischen Veränderungen während dieses Prozesses. Basierend auf einem zuvor etablierten Protokoll7erzeugten wir embryoide Körper (EBs) durch die Hanging-Drop-Technik, gefolgt von induktion unter Verwendung supraphysiologischer Mengen an Retinsäure (RA), um neuronale Vorläuferzellen (NPCs) zu erzeugen, die anschließend zu Neuronen mit neuronalem Induktionsmedium differenziert wurden. Um die Effizienz der Differenzierung zu untersuchen, haben wir zusätzlich zu den traditionellen RT-qPCR- und Immunfluoreszenz(IF)-Assays RNA-seq und Durchflusszytometrie durchgeführt. Diese Analysen liefern eine umfassende Messung des Verlaufs der stufenspezifischen Differenzierung.

Wir beschreiben das schrittweise Verfahren zur Kultivierung und Differenzierung embryonaler Stammzellen von Mäusen in neuronale Linien, gefolgt von einer Reihe von Assays zur Charakterisierung der differenzierten Zellen. Die embryonalen Stammzellen der E14-Maus wurden verwendet, um embryoide Körper durch die Hängende-Drop-Methode zu bilden, und dann induziert, sich durch Retinsäure in neuronale Vorläuferzellen zu differenzieren und schließlich in Neuronen zu differenzieren. Schließlich wurde die RNA-seq-Analyse verwendet, um die globalen transkriptomischen Veränderungen zu profilieren. Diese Methoden sind nützlich, um die Beteiligung bestimmter Gene und Signalwege an der Regulierung des Zellidentitätsübergangs während der neuronalen Differenzierung zu analysieren.

Als Darstellung unserer Methode führten wir ein EB-, NPC- und Neuronendifferenzierungsexperiment an E14-Zellen durch. E14-Zellen wurden auf γ-bestrahlten MEFs kultiviert (Abbildung 1A), bis die γ-bestrahlte MEF-Population verdünnt war. Neural Differentiation of Mouse Embryonic Stem Cells in Serum-free Monolayer Culture

The ability to differentiate mouse embryonic stem cells (ESC) to neural progenitors allows the study of the mechanisms controlling neural specification as well as the generation of mature neural cell types for further study. In this protocol we describe a method for the differentiation of ESC to neural progenitors using serum-free, monolayer culture. The method is scalable, efficient and results in production of ~70% neural progenitor cells within 4 - 6 days. It can be applied to ESC from various strains grown under a variety of conditions. Neural progenitors can be allowed to differentiate further into functional neurons and glia or analyzed by microscopy, flow cytometry or molecular techniques. The differentiation process is amenable to time-lapse microscopy and can be combined with the use of reporter lines to monitor the neural specification process. We provide detailed instructions on media preparation and cell density optimization to allow the process to be applied to most ESC lines and a variety of cell culture vessels.

This protocol describes in detail the method for generating neural progenitors from embryonic stem cells using a serum-free monolayer method. These progenitors can be used to derive mature neural cell types or to study the process of neural specification and is amenable to multiwell format scaling for compound screening.

Analysis of Retinoic Acid-induced Neural Differentiation of Mouse Embryonic Stem Cells in Two and Three-dimensional Embryoid Bodies

Mouse embryonic stem cells (ESCs) isolated from the inner mass of the blastocyst (typically at day E3.5), can be used as in vitro model system for studying early embryonic development. In the absence of leukemia inhibitory factor (LIF), ESCs differentiate by default into neural precursor cells. They can be amassed into a three dimensional (3D) spherical aggregate termed embryoid body (EB) due to its similarity to the early stage embryo. EBs can be seeded on fibronectin-coated coverslips, where they expand by growing two dimensional (2D) extensions, or implanted in 3D collagen matrices where they continue growing as spheroids, and differentiate into the three germ layers: endodermal, mesodermal, and ectodermal. The 3D collagen culture mimics the in vivo environment more closely than the 2D EBs. The 2D EB culture facilitates analysis by immunofluorescence and immunoblotting to track differentiation. We have developed a two-step neural differentiation protocol. In the first step, EBs are generated by the hanging-drop technique, and, simultaneously, are induced to differentiate by exposure to retinoic acid (RA). In the second step, neural differentiation proceeds in a 2D or 3D format in the absence of RA.

Differentiation of Mouse Embryonic Stem Cells into Cortical Interneuron Precursors

GABAergic cortical interneurons are a heterogeneous population of cells that play critical roles in regulating the output of excitatory pyramidal neurons as well as synchronizing the outputs of pyramidal neuron ensembles. Deficits in interneuron function have been implicated in a variety of neuropsychiatric disorders, including schizophrenia, autism, and epilepsy. The derivation of cortical interneurons from embryonic stem cells not only allows for the study of their development and function, but provides insight into the molecular mechanisms underlying the pathogenesis of cortical interneuron-related disorders. Interneurons also have the remarkable capacity to survive, migrate, and integrate into host cortical circuitry post-transplantation, making them ideal candidates for use in cell-based therapies. Here, we present a scalable, highly efficient, modified embryoid body-to-monolayer method for the derivation of Nkx2.1-expressing interneuron progenitors and their progeny from mouse embryonic stem cells (mESCs). Using a Nkx2.1::mCherry:Lhx6::GFP dual reporter mESC line, Nkx2.1 progenitors or their Lhx6-expressing post-mitotic progeny can be isolated via fluorescence-activated cell sorting (FACS) and subsequently used in a number of downstream applications. We also provide methods to enrich for parvalbumin (PV) or somatostatin (SST) interneuron subgroups, which may be helpful for studying aspects of fate determination or for use in therapeutic applications that would benefit from interneuron subgroup-enriched transplantations.

This protocol describes a method for generating cortical interneuron progenitors and post-mitotic interneuron precursors from mouse embryonic stem cells using a modified embryoid body-to-monolayer method. These progenitors/precursors can be used in vitro or fluorescently sorted and transplanted into neonatal neocortex for studying fate determination, or used in therapeutic applications.

Efficient Differentiation of Mouse Embryonic Stem Cells into Motor Neurons

Direct differentiation of embryonic stem (ES) cells into functional motor neurons represents a promising resource to study disease mechanisms, to screen new drug compounds, and to develop new therapies for motor neuron diseases such as spinal muscular atrophy (SMA) and amyotrophic lateral sclerosis (ALS). Many current protocols use a combination of retinoic acid (RA) and sonic hedgehog (Shh) to differentiate mouse embryonic stem (mES) cells into motor neurons1-4. However, the differentiation efficiency of mES cells into motor neurons has only met with moderate success. We have developed a two-step differentiation protocol5 that significantly improves the differentiation efficiency compared with currently established protocols. The first step is to enhance the neuralization process by adding Noggin and fibroblast growth factors (FGFs). Noggin is a bone morphogenetic protein (BMP) antagonist and is implicated in neural induction according to the default model of neurogenesis and results in the formation of anterior neural patterning6. FGF signaling acts synergistically with Noggin in inducing neural tissue formation by promoting a posterior neural identity7-9. In this step, mES cells were primed with Noggin, bFGF, and FGF-8 for two days to promote differentiation towards neural lineages. The second step is to induce motor neuron specification. Noggin/FGFs exposed mES cells were incubated with RA and a Shh agonist, Smoothened agonist (SAG), for another 5 days to facilitate motor neuron generation. To monitor the differentiation of mESs into motor neurons, we used an ES cell line derived from a transgenic mouse expressing eGFP under the control of the motor neuron specific promoter Hb91. Using this robust protocol, we achieved 51&plusmn;0.8% of differentiation efficiency (n = 3; p &lt; 0.01, Student's t-test)5. Results from immunofluorescent staining showed that GFP+ cells express the motor neuron specific markers, Islet-1 and choline acetyltransferase (ChAT). Our two-step differentiation protocol provides an efficient way to differentiate mES cells into spinal motor neurons.

We developed a new protocol to improve efficiency of in vitro differentiation of mouse embryonic stem cells into motor neurons. The differentiated ES cells acquired motor neurons features as evidenced by expression of neuronal and motor neuron markers using immunohistochemical techniques.

Isolation and Culture of Embryonic Mouse Neural Stem Cells

Neural stem cells (NSCs) are multipotent and can give rise to the three major cell types of the central nervous system (CNS). In vitro culture and expansion of NSCs provide a suitable source of cells for neuroscientists to study the function of neurons and glial cells along with their interactions. There are several reported techniques for the isolation of neural stem cells from adult or embryo mammalian brains. During the microsurgical operation to isolate NSCs from different regions of the embryonic CNS, it is very important to reduce the damage to the brain cells to obtain the highest ratio of live and expandable stem cells. A possible technique for stress reduction during isolation of these cells from the mouse embryo brain is the reduction of surgical time. Here, we demonstrate a developed technique for rapid isolation of these cells from the E13 mouse embryo ganglionic eminence. Surgical procedures include harvesting E13 mouse embryos from the uterus, cutting the frontal fontanelle of the embryo with a bent needle tip, extracting the brain from the skull, microdissection of the isolated brain to harvest the ganglionic eminence, dissociation of the harvested tissue in NSC medium to gain a single cell suspension, and finally plating cells in suspension culture to generate neurospheres.

Neuronal Differentiation from Mouse Embryonic Stem Cells In vitro

The neural differentiation of mouse embryonic stem cells (mESCs) is a potential tool for elucidating the key mechanisms involved in neurogenesis and potentially aid in regenerative medicine. Here, we established an efficient and low cost method for neuronal differentiation from mESCs in vitro, using the strategy of combinatorial screening. Under the conditions defined here, the 2-day embryoid body formation + 6-day retinoic acid induction protocol permits fast and efficient differentiation from mESCs into neural precursor cells (NPCs), as seen by the formation of well-stacked and neurite-like A2lox and 129 derivatives that are Nestin positive. The healthy state of embryoid bodies and the timepoint at which retinoic acid (RA) is applied, as well as the RA concentrations, are critical in the process. In the subsequent differentiation from NPCs into neurons, N2B27 medium II (supplemented by Neurobasal medium) could better support the long term maintenance and maturation of neuronal cells. The presented method is highly efficiency, low cost and easy to operate, and can be a powerful tool for neurobiology and developmental biology research.

Here, we established a low cost and easy to operate method that directs fast and efficient differentiation from embryonic stem cells into neurons. This method is suitable for popularization among laboratories and can be a useful tool for neurological research.
