1.成長システム:屋外オープンレースウェイ池の設定
<ol><li>煙道ガス源(8~10%CO2を含む)の近くにオープンな軌道池を設置する。池の原子炉の場所で水と電気が利用可能であり、原子炉が一日の大半を日陰にないことを確認します(図1)。</li>
	<li>燃焼後プロセスでは、煙道ガスが煙突に入って大気中に排出される数メートル前に、0.95cmの燃料ホースを使用して煙道ガスを捕捉します(図2)。</li>
	<li>スタックとコンプレッサーの間に20 Lのウォータートラップとコンデンサー(コイル長~12 m)を使用して、煙道ガスから水を除去します(図2)。 注:煙道ガスは、通常、約9〜1213.8%の水28を含む。また、凝縮器及びパイプラインは排煙16を冷却する。</li>
	<li>藻類の成長を監視するために、以下のセンサをデータロガーに接続する:(1)2つの波長(650および750nm)での吸光度を測定し、1.05g / Lの最大藻類細胞濃度を検出できるリアルタイム光学濃度センサ29;(2) DOセンサ;(3)空気および池の熱電対;(4)pHセンサ;(5)ECセンサ。 メモ:さらに、pHセンサーとECセンサーは送信機に接続されています。データロガーユニットの構成を 図3に示す。</li>

</ol>2. pH制御システム
<ol><li>図2および図3(補足資料A)に示すように、コンプレッサ、制御バルブシステム、およびデータロガープログラムを使用して、煙道ガス噴射を管理します。</li>
	<li>チューブを使用して、制御バルブから石のディフューザーを介して軌道池の底に煙道ガスを導きます。</li>
	<li>pHに基づいて煙道ガスを成長システムに注入する。pH値が8.05より大きい場合、システムは煙道ガスを注入し、pHが8.00未満の場合、システムは成長のない期間に煙道ガス注入を停止します。流量は、標準リットル/分(SLPM)で測定されます。 メモ:制御バルブでは、入口煙道ガス圧力は最大50psiに制限されています。</li>
</ol>3. 藻類の選択とひずみ維持(光と温度)
注:緑藻類クロレラ・ソロキニアナDOE 1412は、Juergen Polle(ブルックリン大学)30,31によって単離され、National Alliance for Advanced Biofuels and Bioproducts(NAABB)によって選択されました。その選択は、Huesemann et al.32,33によって行われた以前の株特性評価研究に基づいていた。屋外のオープンレースウェイ池を使用する場合の南西部地域の藻類スクリーニング、バイオマス生産性、気候シミュレーション培養(温度や光など)に関する研究は、このプロジェクトで使用される方法に情報を提供しました。
<ol><li>培養物を室温(25°C)で12時間/12時間の明暗サイクルを用いて維持する。</li>
	<li>プレート上および小型液体培養(50 mL ~ 500 mL)で増殖させた培養維持のために、光強度を200 μM/m2/sに保ちます。</li>
	<li>液体培養物50 mL~500 mLでは400 μM/m 2/sでスケールアップ増殖し、液体培養では5 L~20 Lで600 μM/m2/sで光強度を維持します。</li>
</ol>4. スケールアップと品質管理
<ol><li>BG11培養液をイオン交換水と以下の塩類を用いて、主要栄養素としてg/Lで調製する:1.5 NaNO3、0.04 K 2 HPO 4、0.075 MgSO 4*H2 O、0.036 CaCl 2*H2O、0.006 (NH 4)5Fe(C6H 4O7)2、0.006 Na 2 EDTA*2H 2 O、 02Na2CO3;2.86 H3 BO 3、1.81 MnCl 2*4H 2O、0.22 ZnSO 4*7H 2 O、0.39 Na 2 MoO 4*2H2 O、0.079CuSO 4*5H 2 O、0.0494 Co(NO3)2*6H2Oの微量栄養素を含む微量元素溶液を1mL/L加える。 注:プレート接種および/または長期保存のために、7.5g / Lのバクト寒天を加える。培養接種のために、寒天の添加は必要ない。オートクレーブ内の培養液を121°Cで21分間滅菌する。</li>
	<li>BG11培地を寒天とともに滅菌層流フードまたはバイオセーフティキャビネット内のペトリ皿に注ぎます。プレートが固く冷たくなったら、再懸濁した凍結藻類ストック培養物から500μLのピペットをピペットし、アンピシリン(100μg/mL)を加える。藻類プレートをシェーカーテーブル(120rpm)で1〜2週間インキュベートする。</li>
	<li>滅菌ループを使用して、培養プレートから単一の藻類コロニーを選択し、クリーンなバイオセーフティキャビネット内の滅菌増殖培地を含む50mLチューブに接種します。小さな液体培養物をシェーカーテーブル(120rpm)上で1週間成長させる。</li>
	<li>50 mLの藻類培養物(線状成長期、OD750nm ≥1)を500 mL液体培地を含む1 Lフラスコに移す。各フラスコにゴム栓とステンレスチューブをはめ込み、通気を提供します。0.2μmの空気滅菌フィルターを使用して空気をろ過します。文化を1〜2週間成長させましょう。分光光度計(OD750nm)を用いて細胞密度をモニターする。</li>
	<li>500 mL の液体培養物を、8 L の非滅菌培養液を含む 10 L のカーボーイに入れ、5%CO2 と 95% の空気の混合物を注入します。次いで、ステップ4.4と同じ条件で藻類を培養する。</li>
	<li>ストックプレートおよび液体培養物(ステップ4.2〜20124.5)を週に1回監視する。アリコートを取り、顕微鏡下で10倍および40倍の倍率で観察し、所望の株の成長を確実にする。文化が侵害されるか、実験に使用されるまで保持しました。汚染された培養物は廃棄する。</li>
</ol>5. 開放池耕作のための濃縮培地調製
<ol><li>微量元素溶液を調製し、1Lメスフラスコに蒸留水(DW)を部分的に充填した。マグネチックスターラーバーを挿入し、 表1 に示す薬品を順次加える。次の成分を添加する前に、各成分が溶解することを確認してください。磁石を取り外し、フラスコを1Lの体積マークまで満たします。</li>
	<li>1Lのガラス瓶にDWを部分的に充填し、マグネチックスターバーを挿入します。容器をマグネチックスターラープレートの上に置き、反応器の最終容量の化学物質を加え、それらを順番に添加して、それぞれが完全に溶解するようにします。 表 2 に、1 L の培地を準備するための化学物質がリストされているので、すべての値に反応器の最終容積を掛けます。ガラス瓶を1Lに充填する。</li>
</ol>6. 屋外オープンレースウェイ池接種
<ol><li>各接種前および収穫後に30%の漂白剤を使用して反応器を徹底的に洗浄する。漂白剤は一晩放置することをお勧めします。反応器をよくすすぎ、すべての漂白剤を取り除きます。</li>
	<li>藻類接種前に、対応する校正手順に従ってすべてのセンサーを校正します。</li>
	<li>軌道池を最大80%まで充填することによって、水源を使用して濃縮培地を希釈する(ステップ5で)。</li>
	<li>藻類で満たされた10Lカーボーイ(線状成長期OD750nm > 2)を用いて反応器に接種し、それを最終体積にする。</li>
	<li>微細藻類を木製パレットで部分的に遮光して~3日間(図4)順応させ、指数関数的な段階が経過したら、光阻害を回避する適応戦略として。 注:この期間はまた、微細藻類が煙道ガスの直接注入によって引き起こされるストレスに適応する時間を提供する。</li>
</ol>7. 発電ステーションでのバッチ成長実験
<ol><li>水の蒸発、パドルホイールモーター、センサー機能など、日常的な変動を検査して記録します。</li>
	<li>コンプレッサーとウォータートラップを毎日排水して検査し、煙道ガスは非常に腐食性が高いため、腐食を最小限に抑えるために余分な水を除去します34。</li>
	<li>各センサー測定値を10秒ごとにスキャンし、10分ごとに平均データを保存するようにデータロガーを構成します。これらには、DO、pH、EC、リアルタイム光学密度、空気および反応器温度が含まれます。</li>
</ol>8. 離散サンプリングとモニタリング
<ol><li>原子炉の最終体積で水位が一定であることを確認してください、さもなければ光学濃度測定は影響を受けるでしょう。</li>
	<li>反応器内の水を補充した後、紫外可視分光光度計を用いて光学密度(540、680、および750nm)による細胞塊測定のために5mLのサンプルを採取する。このプロセスを毎日繰り返します。</li>
	<li>500mLのサンプルを週に3回採取し、顕微鏡観察と無灰乾燥重量(AFDW)に基づくバイオマス濃度を測定します。<ol>
<li>10倍および40倍の対物レンズで顕微鏡観察を行います。さらに、これらの顕微鏡倍率は、ステップ4.6で説明した藻類品質管理の一部として使用されます。</li>
		<li>AFDWにはステップ8.3で400mLのサンプルを使用する<ol>
<li>孔径0.7μmのガラスマイクロファイバーフィルターをアルミ箔トレイにセットし、540°Cで4時間の炉でアルミ箔トレイ/フィルターを前処理します。</li>
			<li>各アルミ箔トレイに#2鉛筆でラベルを付け、その重量(A)を記録し、真空フィルター装置に入れます。</li>
			<li>ろ過する体積を測定する前に、藻類サンプルを激しく攪拌する。十分な藻類サンプルをろ過して、8〜16mgの灰の前/後重量差を与える。実験の過程で使用する重量差を選択し、この値を一定に保ちます。</li>
			<li>藻類サンプルを含む各フィルターをその箔トレイに入れ、105°Cのオーブンに少なくとも12時間置く。</li>
			<li>ホイルトレイ/フィルターを乾燥炉から取り出し、ガラス製デシケーターに入れて水分の取り込みを防ぎます。各ホイルトレイ/フィルター重量(B)を記録します。</li>

	<li>溶媒を用いたマイクロ波支援抽出(MAE)脂質抽出分析のために収穫する前に、2 Lの藻類を保持する。<ol>
<li>藻類サンプルを相対遠心力(RFC)で4,400 x g で15分間遠心分離します。藻類ペレットを取り、80°Cのオーブンを用いて少なくとも24時間乾燥させる。</li>
		<li>藻類サンプルを粉砕し、藻類粉末を計量する(推奨バイオマスの範囲は0.3g〜0.5g)。</li>
		<li>藻類粉末(乾燥藻類バイオマス)をマイクロ波加速反応系(MARS)Xpress容器に加え、フードの下にクロロホルム:メタノール(2:1、v/v)溶媒溶液10mLを加え、容器を閉じ、一晩放置する。</li>
		<li>溶媒センサーを使用して容器をMARSマシンに70°C、800Wの電力で60分間置きます。</li>
		<li>MARSから容器を取り出し、ボンネットの下で冷やします。</li>

</ol>9. 藻類の収穫と輪作
<ol><li>培養物が定常期に達するのに近づいたときに全藻類培養体積の75%を収穫する。実験室でバイオマス生産性分析を行うために2〜20125Lの培養物を取る。残りの藻類を処理し、所望の藻類製品に変換する。</li>
	<li>残りの25%の藻類を接種源として使用して、開いた軌道池を再成長させる。反応器の全容積の80%まで水を加え、濃縮媒体を加えてから、必要に応じて反応器の最終容積まで充填を終える。</li>
	<li>温度と光強度条件に基づいて、季節に応じて適切な藻類株を栽培する。</li>
</ol>10. データ管理
<ol><li>データロガーにデータを記録し、ステップ7.3のように分析のために収集します。</li>

微細藻類は、開放軌道池および閉鎖型フォトバイオリアクター13、14、15、16、17、18、19を含む様々な栽培システムまたは反応器において増殖および生物学的変換を達成することができる。研究者らは、屋内または屋外の条件下で、両方の栽培システムにおけるバイオプロセスの成功を決定する利点と限界を研究しました5,6,16,20,21,22,23,24,25.オープンレースウェイ池は、煙道ガスをスタックから直接分配できる状況での炭素回収と利用のための最も一般的な栽培システムです。このタイプの栽培システムは、比較的安価であり、スケールアップが容易であり、エネルギーコストが低く、混合のためのエネルギー要件が低い。さらに、これらのシステムは発電所と簡単に共存できるため、CCUプロセスをより効率的に行うことができます。しかし、CO2気体/液体物質移動の制限など、考慮する必要があるいくつかの欠点があります。制限はあるものの、屋外の微細藻類バイオ燃料生産に最も適したシステムとして、開放的な軌道池が提案されている5,9,11,16,20。

Methods for Performing Crosses in Setaria viridis, a New Model System for the Grasses

Setaria viridis is an emerging model system for C4 grasses. It is closely related to the bioenergy feed stock switchgrass and the grain crop foxtail millet. Recently, the 510 Mb genome of foxtail millet, S. italica, has been sequenced 1,2 and a 25x coverage genome sequence of the weedy relative S. viridis is in progress. S. viridis has a number of characteristics that make it a potentially excellent model genetic system including a rapid generation time, small stature, simple growth requirements, prolific seed production 3 and developed systems for both transient and stable transformation 4. However, the genetics of S. viridis is largely unexplored, in part, due to the lack of detailed methods for performing crosses. To date, no standard protocol has been adopted that will permit rapid production of seeds from controlled crosses.
The protocol presented here is optimized for performing genetic crosses in S. viridis, accession A10.1. We have employed a simple heat treatment with warm water for emasculation after pruning the panicle to retain 20-30 florets and labeling of flowers to eliminate seeds resulting from newly developed flowers after emasculation. After testing a series of heat treatments at permissive temperatures and varying the duration of dipping, we have established an optimum temperature and time range of 48 &deg;C for 3-6 min. By using this method, a minimum of 15 crosses can be performed by a single worker per day and an average of 3-5 outcross progeny per panicle can be recovered. Therefore, an average of 45-75 outcross progeny can be produced by one person in a single day. Broad implementation of this technique will facilitate the development of recombinant inbred line populations of S. viridis X S. viridis or S. viridis X S. italica, mapping mutations through bulk segregant analysis and creating higher order mutants for genetic analysis.

Methods for Performing Crosses in Setaria viridis, a New Model System for the Grasses

We have developed a methodology for performing crosses in Setaria viridis (S. viridis). The method involves pruning the panicle prior to a hot water treatment to kill viable pollen. Crosses are performed following a well-controlled growth regime and typically result in the recovery of 1 to 7 cross-pollinated seed/s per panicle.

Analysis of Fatty Acid Content and Composition in Microalgae

A method to determine the content and composition of total fatty acids present in microalgae is described. Fatty acids are a major constituent of microalgal biomass. These fatty acids can be present in different acyl-lipid classes. Especially the fatty acids present in triacylglycerol (TAG) are of commercial interest, because they can be used for production of transportation fuels, bulk chemicals, nutraceuticals (&omega;-3 fatty acids), and food commodities. To develop commercial applications, reliable analytical methods for quantification of fatty acid content and composition are needed. Microalgae are single cells surrounded by a rigid cell wall. A fatty acid analysis method should provide sufficient cell disruption to liberate all acyl lipids and the extraction procedure used should be able to extract all acyl lipid classes.
With the method presented here all fatty acids present in microalgae can be accurately and reproducibly identified and quantified using small amounts of sample (5 mg) independent of their chain length, degree of unsaturation, or the lipid class they are part of.
This method does not provide information about the relative abundance of different lipid classes, but can be extended to separate lipid classes from each other.
The method is based on a sequence of mechanical cell disruption, solvent based lipid extraction, transesterification of fatty acids to fatty acid methyl esters (FAMEs), and quantification and identification of FAMEs using gas chromatography (GC-FID). A TAG internal standard (tripentadecanoin) is added prior to the analytical procedure to correct for losses during extraction and incomplete transesterification.

A method for the determination of fatty acid content and composition in microalgae based on mechanical cell disruption, solvent based lipid extraction, transesterification, and quantification and identification of fatty acids using gas chromatography is described. A tripentadecanoin internal standard is used to compensate for the possible losses during extraction and incomplete transesterification.

Optimize Flue Gas Settings to Promote Microalgae Growth in Photobioreactors via Computer Simulations

Flue gas from power plants can promote algal cultivation and reduce greenhouse gas emissions1. Microalgae not only capture solar energy more efficiently than plants3, but also synthesize advanced biofuels2-4. Generally, atmospheric CO2 is not a sufficient source for supporting maximal algal growth5. On the other hand, the high concentrations of CO2 in industrial exhaust gases have adverse effects on algal physiology. Consequently, both cultivation conditions (such as nutrients and light) and the control of the flue gas flow into the photo-bioreactors are important to develop an efficient &ldquo;flue gas to algae&rdquo; system. Researchers have proposed different photobioreactor configurations4,6 and cultivation strategies7,8 with flue gas. Here, we present a protocol that demonstrates how to use models to predict the microalgal growth in response to flue gas settings. We perform both experimental illustration and model simulations to determine the favorable conditions for algal growth with flue gas. We develop a Monod-based model coupled with mass transfer and light intensity equations to simulate the microalgal growth in a homogenous photo-bioreactor. The model simulation compares algal growth and flue gas consumptions under different flue-gas settings. The model illustrates: 1) how algal growth is influenced by different volumetric mass transfer coefficients of CO2; 2) how we can find optimal CO2 concentration for algal growth via the dynamic optimization approach (DOA); 3) how we can design a rectangular on-off flue gas pulse to promote algal biomass growth and to reduce the usage of flue gas. On the experimental side, we present a protocol for growing Chlorella under the flue gas (generated by natural gas combustion). The experimental results qualitatively validate the model predictions that the high frequency flue gas pulses can significantly improve algal cultivation.

Flue gas from power plants is a cheap CO2 source for algal growth. We have built prototype &#34;flue gas to algal cultivation&#34; systems and described how to scale up the algal cultivation process. We have demonstrated the use of a mass-transfer bio-reaction model to simulate and to design the optimal operation of flue gas for the growth of Chlorella sp. in algal photo-bioreactors.

Quantification of Heavy Metals and Other Inorganic Contaminants on the Productivity of Microalgae

Increasing demand for renewable fuels has researchers investigating the feasibility of alternative feedstocks, such as microalgae. Inherent advantages include high potential yield, use of non-arable land and integration with waste streams. The nutrient requirements of a large-scale microalgae production system will require the coupling of cultivation systems with industrial waste resources, such as carbon dioxide from flue gas and nutrients from wastewater. Inorganic contaminants present in these wastes can potentially lead to bioaccumulation in microalgal biomass negatively impact productivity and limiting end use. This study focuses on the experimental evaluation of the impact and the fate of 14 inorganic contaminants (As, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb, Sb, Se, Sn, V and Zn) on Nannochloropsis salina growth. Microalgae were cultivated in photobioreactors illuminated at 984 &#181;mol m-2 sec-1 and maintained at pH 7 in a growth media polluted with inorganic contaminants at levels expected based on the composition found in commercial coal flue gas systems. Contaminants present in the biomass and the medium at the end of a 7 day growth period were analytically quantified through cold vapor atomic absorption spectrometry for Hg and through inductively coupled plasma mass spectrometry for As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Sb, Se, Sn, V and Zn. Results show N. salina is a sensitive strain to the multi-metal environment with a statistical decrease in biomass yieldwith the introduction of these contaminants. The techniques presented here are adequate for quantifying algal growth and determining the fate of inorganic contaminants.

Integration of microalgal cultivation with industrial flue gas will ultimately introduce heavy metals and other inorganic compounds into the growth media. This study presents a procedure used to determine the end fate and impact of heavy metals and inorganic contaminants on the growth of Nannochloropsis salina grown in photobioreactors.

Comparison of Scale in a Photosynthetic Reactor System for Algal Remediation of Wastewater

An experimental methodology is presented to compare the performance of two different sized reactors designed for wastewater treatment. In this study, ammonia removal, nitrogen removal and algal growth are compared over an 8-week period in paired sets of small (100 L) and large (1,000 L) reactors designed for algal remediation of landfill wastewater. Contents of the small and large scale reactors were mixed before the beginning of each weekly testing interval to maintain equivalent initial conditions across the two scales. System characteristics, including surface area to volume ratio, retention time, biomass density, and wastewater feed concentrations, can be adjusted to better equalize conditions occurring at both scales. During the short 8-week representative time period, starting ammonia and total nitrogen concentrations ranged from 3.1-14 mg NH3-N/L, and 8.1-20.1 mg N/L, respectively. The performance of the treatment system was evaluated based on its ability to remove ammonia and total nitrogen and to produce algal biomass. Mean &#177; standard deviation of ammonia removal, total nitrogen removal and biomass growth rates were 0.95&#177;0.3 mg NH3-N/L/day, 0.89&#177;0.3 mg N/L/day, and 0.02&#177;0.03 g biomass/L/day, respectively. All vessels showed a positive relationship between the initial ammonia concentration and ammonia removal rate (R2=0.76). Comparison of process efficiencies and production values measured in reactors of different scale may be useful in determining if lab-scale experimental data is appropriate for prediction of commercial-scale production values.

Cultivation of Green Microalgae in Bubble Column Photobioreactors and an Assay for Neutral Lipids

There is significant interest in the study of microalgae for engineering applications such as the production of biofuels, high value products, and for the treatment of wastes. As most new research efforts begin at laboratory scale, there is a need for cost-effective methods for culturing microalgae in a reproducible manner. Here, we communicate an effective approach to culture microalgae in laboratory-scale photobioreactors, and to measure the growth and neutral lipid content of that algae. Instructions are also included on how to set up the photobioreactor system. Although the example organisms are species of Chlorella and Auxenochlorella, this system can be adapted to cultivate a wide range of microalgae, including co-cultures of algae with non-algae species. Stock cultures are first grown in bottles to produce inoculum for the photobioreactor system. Algae inoculum is concentrated and transferred to photobioreactors for cultivation in batch mode. Samples are collected daily for the optical density readings. At the end of the batch culture, cells are harvested by centrifuge, washed, and freeze dried to obtain a final dry weight concentration. The final dry weight concentration is used to create a correlation between the optical density and the dry weight concentration. A modified Folch method is subsequently used to extract total lipids from the freeze-dried biomass and the extract is assayed for its neutral lipid content using a microplate assay. This assay has been published previously but protocol steps were included here to highlight critical steps in the procedure where errors frequently occur. The bioreactor system described here fills a niche between simple flask cultivation and fully-controlled commercial bioreactors. Even with only 3-4 biological replicates per treatment, our approach to culturing algae leads to tight standard deviations in the growth and lipid assays.

Here, we present a protocol to construct lab-scale bubble column photobioreactors and use them to culture microalgae. It also provides a method for the determination of the culture growth rate and neutral lipid content.

Microalgae Cultivation and Biomass Quantification in a Bench-Scale Photobioreactor with Corrosive Flue Gases

Photobioreactors are illuminated cultivation systems for experiments on phototrophic microorganisms. These systems provide a sterile environment for microalgal cultivation with temperature, pH, and gas composition and flow rate control. At bench-scale, photobioreactors are advantageous to researchers studying microalgal properties, productivity, and growth optimization. At industrial scales, photobioreactors can maintain product purity and improve production efficiency. The video describes the preparation and use of a bench-scale photobioreactor for microalgal cultivation, including the safe use of corrosive gas inputs, and details relevant biomass measurements and biomass productivity calculations. Specifically, the video illustrates microalgal culture storage and preparation for inoculation, photobioreactor assembly and sterilization, biomass concentration measurements, and a logistic model for microalgal biomass productivity with rate calculations including maximum and overall biomass productivities. Additionally, since there is growing interest in experiments to cultivate microalgae using simulated or real waste gas emissions, the video will cover the photobioreactor equipment adaptations necessary to work with corrosive gases and discuss safe sampling in such scenarios.

Bench-scale, axenic cultivation facilitates microalgal characterization and productivity optimization before subsequent process scale-up. Photobioreactors provide the necessary control for reliable and reproducible microalgal experiments and can be adapted to safely cultivate microalgae with the corrosive gases (CO2, SO2, NO2) from municipal or industrial combustion emissions.

Biogas Purification through the use of a Microalgae-Bacterial System in Semi-Industrial High Rate Algal Ponds

In recent years, a number of technologies have emerged to purify biogas into biomethane. This purification entails a reduction in the concentration of polluting gases such as carbon dioxide and hydrogen sulfide to increase the content of methane. In this study, we used a microalgal cultivation technology to treat and purify biogas produced from organic waste from the swine industry to obtain ready-to-use biomethane. For cultivation and purification, two 22.2 m3 open-pond photobioreactors coupled with an absorption-desorption column system were set up in San Juan de los Lagos, Mexico. Several recirculation liquid/biogas ratios (L/G) were tested to obtain the highest removal efficiencies; other parameters, such as pH, dissolved oxygen (DO), temperature, and biomass growth, were measured. The most efficient L/Gs were 1.6 and 2.5, resulting in a treated biogas effluent with a composition of 6.8%vol and 6.6%vol in CO2, respectively, and removal efficiencies for H2S up to 98.9%, as well as maintaining O2 contamination values of less than 2%vol. We found that pH greatly determines CO2 removal, more so than L/G, during cultivation because of its participation in the photosynthetic process of microalgae and its ability to vary pH when solubilized due to its acidic nature. DO, and temperature oscillated as expected from the light-dark natural cycles of photosynthesis and the time of day, respectively. Biomass growth varied with CO2 and nutrient feeding as well as reactor harvesting; however, the trend remained primed for growth.
